Pelagihabitans pacificus gen. nov., sp. nov., a member of the family Flavobacteriaceae isolated from a deep-sea seamount

Abstract: The Gram-stain-negative, orange-pigmented, non-spore-forming, non-motile, strictly aerobic, rod-shaped bacterial strain, designated TP-CH-4T, was isolated from a seamount near the Yap Trench in the tropical western Pacific. The optimal growth conditions were determined to be at pH 7–8, 25–30 °C and in the presence of 2 % (w/v) NaCl. The major respiratory quinone was MK-6. The polar lipid profile contained phosphatidylethanolamine, two unidentified aminolipids, two unidentified phos… Show more

“…Genomic DNAs were extracted using a bacterial genomic DNA Mini kit (TaKaRa Bio) according to the manufacturer's protocol. The gene encoding 16S rDNA was amplified, cloned and sequenced following a previously described procedure ( Wang, Liu, & Zhang, 2020 ). The evolutionary history was inferred using the Neighbor-Joining method ( Saitou, 1987 ).…”

“…At the time of writing, the family Flavobacteriaceae comprises more than 200 genera (https://lpsn.dsmz.de/family/flavobacteriaceae), while only one type strain has been established in the genus Hwangdonia . Members of the family Flavobacteriaceae contain menaquinone 6 as the solar respiratory quinone, have G+C contents ranging from 32 to 37 mol% [3], and are distributed in diverse marine environments such as deep-sea seamount [4], coastal sediment [5], Antarctic seawater [6], marine plants [7] and animals [8]. Submarine cold seeps [9], representing a specific marine environment, release various hydrocarbons through fluids ranging from marine sediment to sea water [10].…”

“…The family Flavobacteriaceae also includes a rather complex group of halophilic organisms, many of which are psychrophilic. Members of most genera classified in the family Flavobacteriaceae produce light to bright yellow or orange pigments, such as, carotenoid or flexirubin, although nonpigmented taxa and strains do occur [15, 16]. Carotenoid pigments are usually produced by members of marine species while flexirubin pigments are more frequently associated with clinical, freshwater or soil organisms [11, 17].…”
