PEDV-spike-protein-expressing mRNA vaccine protects piglets against PEDV challenge

Yongxiang Zhao,

Xu Song
et al.

Abstract: Porcine epidemic diarrhea virus (PEDV), a swine enteropathogenic coronavirus, causes severe diarrhea in neonatal piglets, which is associated with a high mortality rate. Thus, developing effective and safe vaccines remains a top priority for controlling PEDV infection. Here, we designed two lipid nanoparticle (LNP)-encapsulated mRNA (mRNA-LNP) vaccines encoding either the full-length PEDV spike (S) protein or a multiepitope chimeric spike (Sm) protein. We found that the S mRNA-LNP vaccine was superior to the S… Show more

“…A slightly more distant but mRNA vaccine-based veterinary example targeted porcine epidemic diarrhea virus (PEDV) [57]. PEDV is an enteric coronavirus of pigs and causes significant morbidity and mortality in piglets, representing significant losses in the pig industry [58].…”

“…For example, an mRNA lipid nanoparticle (mRNA-LNP) vaccine created by Li’s group, which contains the complete PEDV spike (S) protein, demonstrated the ability to stimulate strong PEDV-specific immune responses in piglets. This vaccine not only effectively shielded actively immunized piglets from PEDV infection but also imparted passive anti-PEDV immunity to neonatal piglets through the transfer of colostrum-derived antibodies from immunized sows [ 157 ]. mRNA vaccines have shown great efficacy in fighting novel coronavirus infections in humans.…”

“…In pregnant sows, mRNA PED vaccination based on the receptor binding region (RBD) heterodimer generated neutralizing antibody levels comparable to those of the available inactivated vaccines [ 129 ]. Another PEDV vaccine that targets the spike (S) protein of PEDV induces a strong antibody response and antigen-specific T-cell responses in immunized piglets [ 130 ]. Furthermore, an mRNA-vectored Nipah virus (NiV) vaccine expressing soluble G glycoprotein candidates in pigs was developed using lipid nanoparticles.…”
