Pectin Dependent Cell Adhesion Restored by a Mutant Microtubule Organizing Membrane Protein

Abstract: The cellulose- and pectin-rich plant cell wall defines cell structure, mediates defense against pathogens, and facilitates plant cell adhesion. An adhesion mutant screen of Arabidopsis hypocotyls identified a new allele of QUASIMODO2 (QUA2), a gene required for pectin accumulation and whose mutants have reduced pectin content and adhesion defects. A suppressor of qua2 was also isolated and describes a null allele of SABRE (SAB), which encodes a previously described plasma membrane protein required for longitud… Show more

“…Additionally, we investigated whether gosamt1 gosamt2 hypocotyls presented cell adhesion phenotypes using ruthenium red dye. Ruthenium red cannot enter into WT hypocotyls, but it can enter and dye hypocotyls of mutants with cell adhesion defects 34 . We observed gosamt1 gosamt2 hypocotyls were stained whereas WT hypocotyls were not ( Fig.…”

“…These HG-modifying enzymes include pectin methylesterases (PMEs) ( Pelloux et al 2007 ), PME inhibitors (PMEIs) ( Coculo and Lionetti 2022 ), acetyl esterases ( Sénéchal et al 2014 ), and polygalacturonases ( Kim et al 2006 ), which hydrolyze the methyl/acetyl esters and/or the HG backbone, respectively ( Amsbury et al 2016 ; Liu et al 2023 ). Changes in HG methyl esterification and/or HG size are associated with numerous HG functions, including cell:cell adhesion ( Kohorn et al 2021a ), cell elongation/shape/growth ( Daher et al 2018 ; Haas et al 2020 ), and fruit ripening ( Paniagua et al 2014 ).…”
