PEBP2/PEA2 represents a family of transcription factors homologous to the products of the Drosophila runt gene and the human AML1 gene.

Abstract: ABSTRACTcDNAs representing the a subunit of polyomavirus enhancer binding protein 2 (PEBP2; also cafled PEA2) were isolated. The products of the cDNAs are highly homologous to that of Drosophila segmentation gene runt (run) for an N-proximal 128-amino acid region showing 66% identity. The run homology region encompasses the domain capable of binding to a specific nucleotide sequence motif and of dimerizing with the companion 13 subunit. The human AMLI gene related to t(8;21) acute myeloid leukemia also has a r… Show more

“…In order to isolate human cDNAs encoding the Runt domain of PEBP2, we screened the cDNA library prepared from a human T-cell line using, as a probe, the mouse cDNA fragment containing the Runt domain region of PEBP2aA1 (Ogawa et al, 1993b). From this screening, the cDNA clones representing human PEBP2aA, PEBP2aB/AML1 and PEBP2aC were isolated.…”

“…One represented the human counterpart of the mouse major splice form, termed PEBP2aA1, characterized by the highly conserved ®ve amino acid sequence, VWRPY, at the extreme carboxy-terminus of the protein product (Ogawa et al, 1993b). The VWRPY motif is present not only in the major products of all three mammalian Runt domain genes but also in the products of Drosophila runt and lozenge as well as that of C. elegans runt (reviewed by Ito and Bae, 1997, in press).…”
