PDGFRα controls the balance of stromal and adipogenic cells during adipose tissue organogenesis

Abstract: Adipose tissue is distributed in depots throughout the body with specialized roles in energy storage and thermogenesis. PDGFRα is a marker of adipocyte precursors, and increased PDGFRα activity causes adipose tissue fibrosis in adult mice. However, the function of PDGFRα during adipose tissue organogenesis is unknown. Here, by analyzing mice with juxtamembrane or kinase domain point mutations that increase PDGFRα activity (V561D or D842V), we found that PDGFRα activation inhibits embryonic white adipose tissue… Show more

“…These cells, which are positive for platelet-derived growth factor receptor  or , seem to derive from a common progenitor as pre-adipocytes destined to mature into adipocytes. These cells therefore seem to be at the crossroad between healthy and unhealthy adipose tissue development (65,66). Despite the fact that the adiponectin promotor has become the preferred tool for generating adipocyte-specific knockout mice, the aP2-driven model applied in this study has the advantage that it also targets the adipocyte precursors.…”

“…Other signaling molecules that are known to regulate adipogenesis, such as PdgfRa, insulin receptor, and Igf1R, localize to the ciliary membrane upon growth arrest and ciliogenesis/ciliary elongation (25,26,29,(66)(67)(68). There is extensive crosstalk among pathways that are mediated by these molecules (69) and evidence that Rpgrip1l may participate in some of this coordination (70,71).…”

“…Within mouse VIS (perigonadal) adipose depot, TCF21 protein was seen exclusively in cells positive for platelet-derived growth factor receptor ␣, a cell-surface marker of fibroblastadipocyte progenitors (Fig. 1F) (19,20). On the contrary, TCF21 was undetectable in SQ (inguinal) depots isolated from the same animals ( Fig.…”
