PDGF-B gene therapy accelerates bone engineering and oral implant osseointegration

Abstract: Platelet-derived growth factor-BB (PDGF-BB) stimulates repair of healing-impaired chronic wounds such as diabetic ulcers and periodontal lesions. However, limitations in predictability of tissue regeneration occur due in part to transient growth factor bioavailability in vivo. Here, we report that gene delivery of PDGF-B stimulates repair of oral implant extraction socket defects. Alveolar ridge defects were created in rats and were treated at the time of titanium implant installation with a collagen matrix co… Show more

“…Among them, there are fibroblast growth factor-2 (FGF-2), insulin-like growth factor (IGF), platelet-derived growth factor (PDGF), TGF-b, and vascular endothelial growth factor A (VEGFA). [5][6][7][8][9][10][11] However, little is known about their bone regeneration capacity relative to BMP-2.…”

“…The fat tissue stem cells application can be carried out as promptly as within 24 hours owing to a special method of tissue engineering, and has positive results in healing bone defects 48 . In addition to wound healing, some authors propose the use of growth factors in the treatment of alveolitis 49 , implant osseointegration 50 and alveolar ridge augmentation 51 . There is evidence that the postoperative healing was better and postoperative pain lower in patients treated with growth-factors-rich plazma 52 .…”

“…70 Moreover, it has been implicated in the formation of new bone in several in vivo studies. [71][72][73][74][75][76] Interestingly, fully differentiated macrophages cultured on Fg also evidenced enhanced secretion of angiogenic-associated proteins. This suggests that when triggered by Fg, macrophages influence different key steps, are able to modulate the inflammatory response, and also contribute to new bone formation and angiogenesis, both of which were stimulated with Fg-adsorbed Ch scaffolds in vivo.…”
