PDE5 is converted to an activated state upon cGMP binding to the GAF A domain

Abstract: cGMP-speci®c, cGMP-binding phosphodiesterase (PDE5) regulates such physiological processes as smooth muscle relaxation and neuronal survival. PDE5 contains two N-terminal domains (GAF A and GAF B), but the functional roles of these domains have not been determined. Here we show that recombinant PDE5 is activated directly upon cGMP binding to the GAF A domain, and this effect does not require PDE5 phosphorylation. PDE5 exhibited time-and concentration-dependent reversible activation in response to cGMP, with th… Show more

“…PDE5 activity is not fixed, but it becomes enhanced when cGMP binds to the enzyme (17). This enhancement corresponds to a time-dependent increase in the limiting rate of hydrolysis (V p ), a reduction in the K m for cGMP, and an increase in the affinity of sildenafil (K i ).…”

“…The equations assume that the enhancement is irreversible, which, considering the short time intervals being analyzed, is justified (17,18). With values assigned to the variables, the cGMP profile is obtained by numerically solving the following equation.…”

“…3 For PDE5, enzyme activation most likely proceeds via phosphorylation of the catalytic subunits and allosteric changes in cGMP binding to the GAF domains. 20,21 (While PDE5 has been reported to copurify with the PDE6 g subunit, 22 the physiological significance is uncertain because the g subunit lacks binding or inhibitory activity toward PDE5 in vitro. 23 )…”
