PCTAIRE3: a putative mediator of growth arrest and death induced by CTS-1, a dominant-positive p53-derived synthetic tumor suppressor, in human malignant glioma cells

Abstract: Chimeric tumor suppressor-1 (CTS-1) is based on the sequence of p53 and was designed as a therapeutic tool resisting various mechanisms of p53 inactivation. We previously reported that an adenovirus expressing CTS-1 (Ad-CTS-1) has superior cell deathinducing activity in glioma cells compared with wild-type p53. Here, we used cDNA microarrays to detect changes in gene expression preferentially induced by Ad-CTS-1. The putative serin threonine kinase, PCTAIRE3, and the quinone oxireductase, PIG3, were strongly i… Show more

“…Strong evidence provided by this study and other studies (56,57) suggests that PIG3 apoptotic action is through, or results in, oxidative stress. In this regard we have demonstrated that in vitro PIG3 activity generates ROS, and that PIG3 overexpression in cells leads to ROS generation, which is dependent on active enzyme or at least on protein able to bind cofactor.…”

“…Migrated cells on the lower layer of the membrane were fixed, stained with hematoxylin/eosin and counted. Number of migrated cells was normalized to cell density at the end of the migration period, assessed in parallel by crystal violet staining as previously described (13).…”

“…Lastly, PCTAIRE-3 as well as PCTAIRE-2 have been implicated in Alzheimer's disease [59,60]. CDK18 was induced by CTS-1 (Chimeric tumor suppressor-1, p53derived synthetic tumor suppressor) and mediated growth arrest and death in glioma cells [61]. Apart from its activation by cyclin A2, in the same study CDK18 was shown to phosphorylate retinoblastoma tumorsupressor protein (Rb) in vitro [55].…”
