Terubok (Tenualosa toli) is one of Malaysia commercially important fish which is found mainly in Sarawak. Their numbers of catch have been declining in the past 15 years due to heavy exploitation. Hence, a study was done to determine the mitochondrial genetic diversity of T. toli from Daro and Mukah, Sarawak inferred by partial Cytochrome b gene. DNA extraction was done on 84 T. toli samples. PCR amplification using Cyt-b primers had been carried out and sequence of 469 bp length was obtained from each sample. Next, phylogenetic analysis was performed to study the relationship among the individuals. The genetic diversity of the population was determined through the haplotype and nucleotide diversity. Result of the study showed that the Haplotype Diversity (HD) of T. toli was relatively low for Daro (Hd = 0.232) and Mukah populations (Hd = 0.178). The nucleotide diversity was also low for both populations. Moreover, only nine haplotypes were identified from the 84 individuals. A single haplotype was shared amongst 76 individuals. These findings correlate with previous study that showed the number of the fish caught had declined drastically and might cause a genetic deprivation towards its population. Comparably, maximum likelihood analysis revealed that two T. toli individuals were separated from the main clade, suggesting that these two individuals might come from another Terubok population. Genetic diversity of the mitochondrial DNA Cytochrome b of the T. toli obtained in this study would be useful in the implementation of conservation and fisheries management of this species in Sarawak.

Wild populations of C. marulius have undergone a steady decline due to habitat deterioration, overexploitation and aquatic pollution. Genetic polymorphism by using microsatellite DNA markers was analyzed in natural populations of C. marulius. Samples were collected from different sites of river Jhelum viz. Trimu Headworks, Rasul Barrage, Mangla Dam, Jhelum Bridge, and Marala Dhand. A total of five microsatellite loci (CA05, CA07, CA08, CA09, and CA10) were used to evaluate genetic polymorphism. The overall populations of C. marulius showed a moderate level of heterozygosity. The mean observed heterozygosity (Ho) at all the five loci was 0.43 whereas expected heterozygosity (He) was 0.54. The pairwise population genetic differentiation (FST) ranged in between 0.000001 to 0.231955. Analysis of molecular variance (AMOVA) revealed that majority of variations among populations than within populations 62.05% and 33.03%, respectively. Significant deviation (P<0.05) from Hardy-Weinberg equilibrium was seen in all the populations with clear deficit heterozygosity suggesting inbreeding. A UPGMA tree based on Nei's genetic distance matrix revealed that C. marulius populations were divided into two major clusters. Principle component analysis showed same clustering pattern. The present study about genetic structure of C. marulius populations will be fruitful for effective management and conservation of fisheries resources.

The spotted sardinella, Amblygaster sirm (Walbaum, 1792), is a commercial sardine commonly caught in Malaysia. Lack of management of these marine species in Malaysian waters could lead to overfishing and potentially declining fish stock populations. Therefore, sustainable management of this species is of paramount importance to ensure its longevity. As such, molecular information is vital in determining the A. sirm population structure and management strategy. In the present study, mitochondrial DNA Cytochrome b was sequenced from 10 A. sirm populations: the Andaman Sea (AS) (two), South China Sea (SCS) (six), Sulu Sea (SS) (one), and Celebes Sea (CS) (one). Accordingly, the intra-population haplotype diversity (Hd) was high (0.91–1.00), and nucleotide diversity (π) was low (0.002–0.009), which suggests a population bottleneck followed by rapid population growth. Based on the phylogenetic trees, minimum spanning network (MSN), population pairwise comparison, and FST,and supported by analysis of molecular variance (AMOVA) and spatial analysis of molecular variance (SAMOVA) tests, distinct genetic structures were observed (7.2% to 7.6% genetic divergence) between populations in the SCS and its neighboring waters, versus those in the AS. Furthermore, the results defined A. sirm stock boundaries and evolutionary between the west and east coast (which shares the same waters as western Borneo) of Peninsular Malaysia. In addition, genetic homogeneity was revealed throughout the SCS, SS, and CS based on the non-significant FSTpairwise comparisons. Based on the molecular evidence, separate management strategies may be required for A. sirm of the AS and the SCS, including its neighboring waters.
