PCR‐based gut content analysis of insect predators: using ribosomal ITS‐1 fragments from prey to estimate predation frequency

Abstract: We used polymerase chain reaction to determine whether Ostrinia nubilalis (Hübner) (Lepidoptera: Crambidae) DNA was present in the guts of larvae and adult males and females of the generalist predator Coleomegilla maculata De Geer (Coleoptera: Coccinellidae). The predators were fed Ostrinia nubilalis egg masses and allowed to digest at either 20 degrees C or 27 degrees C for time spans ranging from 0 to 12 h. Four primer pairs, specific for O. nubilalis were developed, using a nuclear ribosomal RNA sequence in… Show more

“…There have also been a few studies using target sequences within multi-copy nuclear ribosomal genes. For example Hoogendoorn and Heimpel (2001) investigated predation of the European corn borer Ostrinia nubilalis (Hu¨bner) (Lepidoptera: Crambidae) by Coleomegilla maculata (DeGeer) (Coleoptera: Coccinellidae) and H. axyridis through targeting a section of the ITS 1 region. Other researchers have rejected the ITS 1 region as it can show high levels of intraspecific variation.…”

“…For example, Hoogendoorn and Heimpel (2001) investigated detectability of four different sized fragments (492, 369, 256 and 150 bp) of the nuclear ribosomal ITS 1 sequence of O. nubilalis within the gut of C. maculata. The shortest (150 bp) fragment was present for the full 12 h of the feeding assay, whereas the two longest fragments disappeared after 4 h and 5 h respectively.…”

“…Recently, gut content analysis using molecular markers have been developed and used for many predatory species (e.g., Agustí et al, 2003;Greenstone, 2003;Hoogendoorn and Heimpel, 2001;Itou et al, 2013;Kamikawa et al, 2016;Symondson, 2002;Wari et al, 2014;Zhang et al, 2007). Phytoseiid mites─especially specialist species─tend to consume spider-mite eggs (Blackwood et al, 2001;Furuichi et al, 2005), and the small size of this preferred prey item hampers detection in the gut.…”

“…Molecular DNA makers have enjoyed wide use for diagnostic purposes within and between species in insects Paskewitz 1996, Hoogendoorn andHeimpel 2001). Polymerase chain reaction (PCR)-based analysis allows assays of small amounts of DNA from individual insects as well as permitting high throughput of large numbers of samples.…”
