PBMC-Derived T Cells

Abstract: T cell cultures are a valuable tool in food research to perform studies within the food allergy fi eld. Their main applications aim to analyze immunological responses towards food protein antigens to gain further insights into the mechanisms responsible for the development of oral tolerance or for the triggering of food allergies. This chapter describes the main applications, isolation techniques, and culture conditions for PBMC-derived T cells. Furthermore, critical parameters of the model, together with the … Show more

“…The strength and polarity of GABA-A R-activated chloride currents depend on the intracellular chloride concentration set by CCCs [ 17 ]. In murine T cells, only NKCC1 has been detected to date, but in human PBMCs (T cells constitute ~ 45–70% of PBMCs), NKCC1, KCC1, KCC3, KCC4 were detected [ 52 , 95 , 96 ]. One single subunit of GABA-B Rs (B1) was detected in human PBMCs and it remains enigmatic if functional GABA-B R homodimers can be formed in T cells [ 95 ].…”

“…T cells, classically described as the main mediators of beta cell destruction in T1D [ 2 ], are the most abundant cell type in peripheral blood mononuclear cells (PBMCs), making up approximately 45–70% [ 3 ]. The study of PBMCs provides important insight into the immunological status of patients with T1D [ 4 ], and circulating leukocytes are sensors of metabolic stress in patients and act as bioenergetic biomarkers [ 5 ].…”

“…T cells comprise 45-70% of isolated mononuclear cells from human peripheral blood 55 . T cells that primarily rely on mitochondrial OXPHOS for ATP generation are resting naïve T cells, antigenspecific memory T cells, and FoxP3+ regulatory T cells, whereas proliferative helper (CD4+) and cytotoxic (CD8+) effector T cells utilize glycolysis to a higher extent [56][57][58][59] .…”
