Pax6 Is Required for Making Specific Subpopulations of Granule and Periglomerular Neurons in the Olfactory Bulb

Abstract: Sey/Sey progenitors produce neuroblasts capable of migrating into the OB but fail to generate dopaminergic periglomerular and superficial granule cells. Interestingly, superficial granule neurons also express mRNA for tyrosine hydroxylase, the rate-limiting enzyme in dopamine synthesis. Our data show that SVZ neuroblasts are heterogeneous and that Pax6 is required in a cell-autonomous manner for the production of cells in the dopaminergic lineage.

“…In contrast, the production of CR-and TH-expressing PG cells follows an opposite trend with a progressive increase with age (De Marchis et al 2007 ). According to previous studies (Hack et al 2005 ;Kohwi et al 2005 ;Waclaw et al 2006 ) this work suggests that the population of SVZ progenitors with a PG fate is composed of a heterogeneous pool of neurochemically specified elements, and indicates that the relative contribution of these different subsets changes with time. Therefore, neurogenesis in the OB does not reflect a simple turnover of the whole interneuron population of the GL, but it may serve to finely modulate the relative amounts of distinct OB interneuron subpopulations.…”

“…Indeed, Pax6 expression in RMS precursor cells is required to progress towards a PG neuron subtype and its target deletion strongly reduces the proportion of PG cells normally generated (Hack et al 2005 ). Moreover, Pax6 governs the specific differentiation of dopaminergic periglomerular and superficial granule cells in the adultOB (Hack et al 2005 ;Kohwi et al 2005 ). Pax6 is expressed in most proliferating SVZ progenitors but only in a subpopulation of migrating neuroblasts, further indicating that migrating neuroblasts are heterogeneous.…”

“…In addition, forced expression of Pax7 in satellite cells prevents their difference into myoblasts (Olguin and Olwin, 2004). Similarly the activity of Pax6 is involved in conferring progenitor status to neuronal populations in the spinal cord, cerebral cortex, adult hippocampus, and olfactory bulb (Grindley et al, 1995;Kohwi et al, 2005;Bel-Vialar et al, 2007;Tucker et al, 2008). Similarly, the expression of Pax2 and Pax8 is associated with proliferating progenitor populations in the developing kidney (Bouchard et al, 2002) and in the sensory patches of the inner ear (Li et al, 2004;Warchol and Richardson, 2009).…”
