SUMMARYComplex patterns and networks of genes coordinate rhombomeric identities, hindbrain segmentation and neuronal differentiation and are responsible for later brainstem functions. Pax6 is a highly conserved transcription factor crucial for neuronal development, yet little is known regarding its early roles during hindbrain segmentation. We show that Pax6 expression is highly dynamic in rhombomeres, suggesting an early function in the hindbrain. Utilization of multiple gain-and loss-of-function approaches in chick and mice revealed that loss of Pax6 disrupts the sharp expression borders of Krox20, Kreisler, Hoxa2, Hoxb1 and EphA and leads to their expansion into adjacent territories, whereas excess Pax6 reduces these expression domains. A mutual negative cross-talk between Pax6 and Krox20 allows these genes to be co-expressed in the hindbrain through regulation of the Krox20-repressor gene Nab1 by Pax6. Rhombomere boundaries are also distorted upon Pax6 manipulations, suggesting a mechanism by which Pax6 acts to set hindbrain segmentation. Finally, FGF signaling acts upstream of the Pax6-Krox20 network to regulate Pax6 segmental expression. This study unravels a novel role for Pax6 in the segmental organization of the early hindbrain and provides new evidence for its significance in regional organization along the central nervous system.

Left-right asymmetries in the epithalamic region of the brain are widespread across vertebrates, but their magnitude and laterality varies among species. Whether these differences reflect independent origins of forebrain asymmetries or taxa-specific diversifications of an ancient vertebrate feature remains unknown. Here we show that the catshark Scyliorhinus canicula and the lampreys Petromyzon marinus and Lampetra planeri exhibit conserved molecular asymmetries between the left and right developing habenulae. Long-term pharmacological treatments in these species show that nodal signalling is essential to their generation, rather than their directionality as in teleosts. Moreover, in contrast to zebrafish, habenular left-right differences are observed in the absence of overt asymmetry of the adjacent pineal field. These data support an ancient origin of epithalamic asymmetry, and suggest that a nodal-dependent asymmetry programme operated in the forebrain of ancestral vertebrates before evolving into a variable trait in bony fish.

PAX6 expression was examined during early development of the diencephalon and midbrain of Alligator using an immunocytochemical methodology. These observations focused on the basal plate to determine whether diencephalic prosomere organization in this region followed a pattern previously identified for alar plate areas. PAX6 expression was also described in alar diencephalic regions and the adjacent midbrain. PAX6 (+) cells in the basal plate were first seen in prosomere 1 at stage 7, in the midbrain at stage 10, and lastly in prosomeres 2 and 3 at stage 11. By stage 12, a nearly continuous column of PAX6 (+) cells extended from the midbrain basal plate through the entire diencephalon. In the diencephalon, PAX6 (+) cells in the basal plate were of greatest number in prosomere 1, least in prosomere 2, and intermediate in prosomere 3. This pattern of PAX6 expression distinguished these individual basal plate prosomeres. These results indicate that basal plate prosomeres follow a pattern similar to alar plate prosomereric organization during the later stages of early diencephalon development. Over a comparable time period of early diencephalon development, similar observations have been made in chick basal plate. In Alligator and chick, PAX6 expression in the basal plate is similar in the midbrain and prosomere 1 but different in prosomeres 2 and 3: present in Alligator and absent in chick. In alar plate areas of the Alligator diencephalon, PAX6 expression follows a similar pattern to that described for chick and mouse. These similarities in PAX6 expression in alar diencephalic prosomeres suggest that this is a common feature of amniotes. Differential PAX6 expression in alar prosomere 1 and the midbrain in Alligator is similar to that described for a wide range of species which suggests that these features are common to all vertebrates.
