Patterns of cancer cell sphere formation in primary cultures of human oral tongue squamous cell carcinoma and neck nodes

Abstract: Recently a sub-population of cells with stem cell characteristics, reported to be associated with initiation, growth, spread and recurrence, has been identified in several solid tumors including oral tongue squamous cell carcinoma (OTSCC). The aim of our pilot study was to isolate CD44+ cancer stem cells from primary cultures of OTSCC and neck node Level I (node-I) biopsies, grow cell spheres and observe their characteristics in primary cultures. Parallel cultures of hyperplastic lesions of tongue (non-cancer)… Show more

“…For example, Saleem et al found that lymph node metastatic OSCC express higher levels of CD44/CD24. 49 It has been reported that downregulation of CD24 inhibits proliferation while inducing apoptosis in malignant cells. 50 Further, CD24 may be a potential marker for cancer stem cells.…”

“…As such, tumor spheres extracted from OSCC cells show higher stem-like features. They show greater volume of expression of pluripotent transcription agents such as Lin28, Nanog, KLF4, Oct4, and Sox2 in comparison to the respective adherent mono-layer cells [26,60,70,71]. Moreover, influential expression of CSC specific markers, including CD44 and ALDH1 is expressed by oral tumor sphere-forming cells [60,61,72].…”

“…Interestingly, CD44+ cells from hyperplastic or non-cancer tongue tissue did not produce spheres. Besides, primary cultures from metastatic TSCC specimens showed stronger CD44/CD24 expression and produced much more spheres in significantly shorter duration compared to those of node negative TSCC and normal tongue specimens (Saleem et al 2014).…”

“…It has been shown to be deregulated in several cancers including TSCC (Lindquist et al 2012, Yanamoto et al 2014. CD44 has important roles during carcinogenesis and metastasis in TSCC and is involved in several carcinogenic processes like proliferation, migration, cellcell interactions, and apoptosis in TSCC cells (Saleem et al 2014. Its expression shows significant correlation with several other critical biomarkers such as CD24 ( The expression of CD44 in TSCC samples was initially investigated in 1996, where 83 head and neck squamous cell carcinoma (including 12 TSCC specimens) samples were used for immunohistochemical analysis of CD44 variants (v5, v6, v7, v7-v8, and v10) in normal, dysplastic squamous epithelia, primary and metastatic squamous cell carcinoma samples.…”
