Pathophysiology of unilateral pulmonary aspergillosis in an experimental rat model

Abstract: Because little is known about the pathophysiology of invasive pulmonary aspergillosis (IPA), we examined changes in pulmonary and general physiology during this disease in an animal model. In a model of fatal left-sided IPA, 19 persistently neutropenic rats were monitored for clinical signs including body temperature, body weight and respiratory distress. A separate group of nine rats with IPA was used for measurements of arterial blood pressure, arterial O2 and CO2 pressure, lung compliance and surfactant fun… Show more

“…In our laboratory, we developed a model of unilateral IPA in rats that closely mimics the human disease in patients with transient neutropenia (8,9,41). This model is characterized by a transiently neutropenic state, fungal inoculation through the respiratory route, fungal broncho-and angioinvasion, and extrapulmonary dissemination.…”

“…The rat model of aerogenic IPA in transiently neutropenic rats was used, as described previously (8,9,41,42). In short, in neutropenic female RP strain albino rats left-sided invasive IPA was induced by instillation of an A. fumigatus conidial suspension, followed by inhalation.…”

“…Injectable glycopeptides and carbapenems were sporadically used to prevent infections due to multi-resistant staphylococci and enterobacteriae (Melchers et al, 1994; Hashimoto et al, 1998; Yonezawa et al, 2000; Benjamim et al, 2005; Steinbach et al, 2006; Tansho et al, 2006; van Vianen et al, 2006; Cramer et al, 2008; Pinchai et al, 2009; Rivera et al, 2009; Petrik et al, 2010, 2012, 2014; Grahl et al, 2011; Martinez et al, 2013; Verwer et al, 2013). Likewise, multi-antibiotic associations were sometimes reported, like the following scheme: ciprofloxacin (660 mg/L) and polymyxin B (100 mg/L) in water, plus IM amoxicillin (40 mg/kg/day) and a single shot of IM gentamicin (6 mg/kg) at time of experimental infection (Leenders et al, 1996; Dams et al, 1999; Becker et al, 2000, 2002a,b, 2003, 2006; Van Etten et al, 2000; Ruijgrok et al, 2001, 2005, 2006). Interestingly, a few authors preconized to dilute halogenated derivatives, like chlorine or iodine, as disinfectants in drinking water (Corbel and Eades, 1977; Xu et al, 2015).…”

“…In spite of recent major advances (Sable et al, 2008; Brown, 2011; Steele and Wormley, 2012; WĂŒthrich et al, 2012; Drew et al, 2013; Lanternier et al, 2013), there are still many concerns to be addressed: for example, why a particular strain is more virulent than another (Becker et al, 2006)? How to prevent a contamination?…”

“…sojae , A. tamarii and A. nomius [18,19]. Sequence-based approaches include analysis of rDNA ITS and 28S rRNA variable regions [20,21], together with calmodulin and ÎČ-tubulin gene regions [7,22,23]. Variability in the latter two genes can be appropriate for resolving closely related Aspergillus species [24].…”
