Pathophysiology of lipid droplet proteins in liver diseases

Abstract: Cytosolic lipid droplets (LDs) are present in most cell types, and consist of a core comprising neutral lipids, mainly triglycerides and sterol esters, surrounded by a monolayer of phospholipids. LDs are heterogeneous in their structure, chemical composition, and tissue distribution. LDs are coated by several proteins, including perilipins and other structural proteins, lipogenic enzymes, lipases and membrane-trafficking proteins. Five proteins of the perilipin (PLIN) family (PLIN1 (perilipin), PLIN2 (adipose … Show more

“…CREBH is a liver-specific transcription factor and a UPR gene dependent on ATF6 [18,47]. Moreover, PLIN1 is normally found in adipocytes and is responsible for the coating of large LDs [48]. It interacts with Fsp27β/CIDEC and mediates the exchange of lipids between LDs [49].…”

“…LDs increase in abundance in many diseases involving disrupted or rewired lipid metabolism, including non-alcoholic fatty liver disease and cancer [11, 83-86]. In murine hepatocytes, the FA composition and degree of saturation in the triglyceride fraction of purified LDs are both very sensitive to nutritional and genetic perturbations [24, 87, 88].…”

“…PLIN3 and PLIN5 Do Not Compensate for Loss of PLIN2 in WD and CD Livers-Because murine livers also express PLIN3 and PLIN5 (20,21), we were interested in determining whether loss of PLIN2 altered their expression. We first examined Plin2, Plin3, and Plin5 gene expression in both WT and Plin2-null mice fed WD or CD for 30 weeks.…”
