In this experiment, we aimed to recorded the effect of chronic oral exposure to DEHP on lipid peroxidation and histopathological lesions induced in different organs. A total of 150 quail chick divided into two groups (75 quail's chicks at each group), control group left without any treatment all time, the second group was orally administrated 5% of DEHP (278.05 mg/kg bw) three times per week and lasted for 60 days. At 30 and 60 days old, a total number of 25 from both groups were euthanized. Blood samples were collected to determine the serum concentration of DEHP and mono ethyl hexyl phthalate (MEHP), liver and kidney to estimate tissue concentration of malondialdehyde (MDA), samples from different organs for histopathological examination. The result showed that the concentration of DEHP and MEHP in blood's serum and concentration of MDA in liver and kidney significantly increased with time in comparison with the control group; besides, oral administration of DEHP induced sever pathological lesions, in brain, aorta, intestines, liver, lung, ovary and testis. In conclusion, the DEHP and its metabolic product MEHP significantly increased in serum and cause significant increase in concertation of MDA in liver and kidney tissues, produces a significant histopathological change in brain, aorta, liver, intestine, lung, ovary and testis that can cause a life-threatening condition.

HE POULTRY industry has become a critical component of the country's economy, and as a result, detecting and diagnosing disease infections before they spread in the herd limits the massive losses that poultry breeders face. This study was to look for serological diagnosis of mycoplasma disease in broiler using enzyme linked immune sorbent assay ELISA, as well as the histopathological changes in some organs. Between November 2021 and March 2022, this study was carried out broiler fields in Nineveh province. One hundred eighty blood samples were collected from 12 broiler fields. Results revealed the total infection rate of the examined fields was 55/180 (30.56%). In addition, percentage of M. gallisepticum infection was highest in the broiler fields located east of the province 21/45 (46.66%), followed by the percentage of infection in the fields located south of the province 18/45 (40%). While the infection rate was lower in the fields on the north and west of the province, 11/45 (24.44%) and 5/45 (11.11%), While the infection rate was lower in the fields to the north and west of the province, 11/45 (24.44%) and 5/45 (11.11%), respectively. Histopathological changes recorded severe tracheitis, severe inflammation in lung with purulent mucoid secretions in the bronchial and between the alveoli. The air sac showing also severe airsaculitis serous fibrin inflammatory secretion in the lumen with inflammatory cell of polymorphnuclear inflammatory cells infiltration and wall thickening. ELISA is a cost-effective tool for diagnosis of M. gallisepticum infection in broiler chickens, with high incidence in Nineveh province.

Histopathology considered as an essential keystone to understanding diseases on a cellular level, without examining affected tissues and cells; will lack the accurate diagnosis. The most common protocol that used worldwide for histopathology is termed as formalinfixed paraffin-embedded techniques; this protocol is the oldest protocol and still used, this technique not invented once, but it is started with a simple process of tissue fixation then undergo several successful steps as it's known today. This protocol depends on formalin to induce tissue fixation and preservation simultaneously, followed by a series of steps to facilitate infiltration of hot paraffin wax in the cells to adjust the density of tissue similar to that of paraffin wax to support tissue during sectioning process at few micrometers of thickness. Many protocols have been invented and modified subsequently to increase the accuracy of tissue processing and facilitate their study under light microscopy. Additionally, the formalin-fixed and paraffin-embedded samples can be considered the most durable process till now. In this review, we are trying to view all fundamental aspects of classical tissue processing that was depended since 1850s, starting from tissue examination, sampling, accessory, labelling, fixation, dehydration, clearance, infiltration, and embedding with hot paraffin wax. Additionally, explain the routine sectioning and staining with classical Hari's hematoxylin and alcoholic eosin staining protocol.
