Pathological impairments induced by interstitial implantation of 125I Seeds in spinal canal of banna mini-pigs

Abstract: BackgroundUse a banna mini-pig to set up 125I implantation model, and investigate the consequence of radiation-related impairments.MethodsIn present study, 125I seeds were implanted into spinal canal of T13 level of spine in banna mini-pigs. After operation, the pigs were raised up to 8 months, behavior changes were recorded within this period. After 8 months, spinal cords were collected for pathological analysis.ResultsIn this study, a 125I brachytherapy animal model had been successfully established, in the … Show more

“…In the study, we also successfully reduced PERK expression by intrathecal administration of the lentivirus vector in radiation myelitis banna pigs [10], [11] caused by 125 I brachytherapy, the apoptosis rates were significantly higher than that in control group and deteriorated radiation myelitis of banna pigs, which was consistent with that in neural cells. In conclusion, autophagy caused by 125 I radiation was mainly as an attempt of cell survival at an early stage, and it would be a self-destructive process and promoted the process of apoptosis and necrosis radiated by 125 I for more than 72 hours.…”

“…In both groups, eight brachytherapy seeds were implanted into the spinal dura mater at the T13 level. Radiation dosimetry was based on previous studies [10], [11]. The average radiation dose for each group was 16.76±0.32Gy and 17.02±0.58Gy.…”

“…Previously, we successfully established an animal model to study the pathological impairments induced by interstitial implantation of 125 I seeds in the spinal canal of banna mini-pigs [8]. In the current study, we used this model to investigate whether percutaneous vertebroplasty combined with interstitial implantation of 125 I seeds in banna mini-pigs could alleviate the pathological impairments.…”

“…Briefly, at the beginning, we calculated the initial dose (termed as D (0) ) immediately after the 125 I particles were implanted into the spinal cord T13 level; the formula was D (0) = A 0 × 1.27 × Λ × g (r) × F (r θ) /r 2 . A 0 is the particle initial radiation dose, which was tested by CRC-15R calculator on the day before implanting; Λ is constant parameter for 125 I, in our research the value is 1.06; r is the distance between the spinal surface to the 125 I particles, which was obtained from the magnetic resonance imaging (MRI) detecting data; g (r) is radial dose functions; and F (r θ) is anisotropy constant; the detailed data we calculated based on the published method [8,9]. After D 0 had been confirmed, the radiation dose received by the spine was calculated by the formula D (T) = D (0) *T 1/2 *1.443*(1- e -T*0.693/T1/2 ).…”
