Objective. Th e aim of the current study was to assess the protective outcome of hemin, a heme oxygenase-1 (HO-1) inducer on L-arginine-induced acute pancreatitis in rats. Acute pancreatitis (AP) is considered to be a critical infl ammatory disorder with a major impact on the patient health. Various theories have been recommended regarding the pathophysiology of AP and associated pulmonary complications.Methods. Twenty-four adult male albino rats were randomly divided into four groups: control group, acute pancreatitis (AP), hemin pre-treated AP group, and hemin post-treated AP group.Results. Administration of hemin before induction of AP signifi cantly attenuated the L-arginine-induced pancreatitis and associated pulmonary complications characterized by the increasing serum levels of amylase, lipase, tumor necrosis factor-α, nitric oxide, and histo-architectural changes in pancreas and lungs as compared to control group. Additionally, pre-treatment with hemin signifi cantly compensated the defi cits in total antioxidant capacities and lowered the elevated malondialdehyde levels observed with AP. On the other hand, post-hemin administration did not show any protection against L-arginine-induced AP.Conclusions. Th e current study indicates that the induction of HO-1 by hemin pre-treatment signifi cantly ameliorated the L-arginine-induced pancreatitis and associated pulmonary complications may be due to its anti-infl ammatory and antioxidant properties.

“…Based on the results of cell cultures, animal models and clinical trials, ample evidence validates that curcumin at the doses of 100 mg/kg may have potential as a therapeutic agent in AP (Gulcubuk et al, 2013; Zhong, 2015). Curcumin’s improvement of AP is achieved through repressing the infiltration of inflammatory cells, inhibiting lipid peroxidation, and regulating TLR-4/NF-κB and PPARγ/NF-κB signaling pathway, preventing free radical injury and the prevalence of bacterial translocation, thereby reducing trypsin activation, oxidative enzymes, and tissue injury (Gulcubuk et al, 2005, 2006, 2013; Yu et al, 2011; Zhong, 2015). Moreover, curcumin suggests that the activation of caspase-3 may lead to an increase in apoptosis in the early and late phases of experimental AP.…”

Acute pancreatitis (AP) is a commonly occurring gastrointestinal disorder. An increase in the annual incidence of AP has been observed, and it causes acute hospitalization and high mortality. The diagnosis and treatment guidelines for AP recommend conservative medical treatments focused on reducing pancreatic secretion and secondary injury, as a primary therapeutic approach. Unfortunately, the existing treatment options have limited impact on the incidence and severity of AP due to the complex and multifaceted pathological process of this disease. In recent decades, Chinese herbal medicines (CHMs) have been used as efficient therapeutic agents to attenuate AP in Asian countries. Despite early cell culture, animal models, and clinical trials, CHMs are capable of interacting with numerous molecular targets participating in the pathogenesis of AP; however, comprehensive, up-to-date communication in this field is not yet available. This review focuses on the pharmacological activities of CHMs against AP in vitro and in vivo and the underlying mechanisms. A computational prediction of few selected and promising plant-derived molecules (emodin, baicalin, resveratrol, curcumin, ligustrazine, and honokiol) to target numerous proteins or networks involved in AP was initially established based on a network pharmacology simulation. Moreover, we also summarized some potential toxic natural products for pancreas in order to more safe and reasonable medication. These breakthrough findings may have important implications for innovative drug research and the future development of treatments for AP.

Background/Aim. Organophosphate pesticides (OPs) are extensively used worldwide in agriculture and forestry, and their application represents a major health problem for humans and animals. The aim of this study is to investigate the possibility of adaptation of organism to prolonged administration of low dose of diazinon. Methods. The study was conducted on a total of 60 male Wistar rats. The first 30 rats was divided into four equal diazinon groups (n=6) and the control one (corn oil). Diazinon was orally administered once at doses: 200, 400, 600, 800 mg/kg (one doseone group). The concentration of glucose, activity of α-amylase and relative activity of LDH1-LDH5 isoenzymes in the blood were measured 24 hours after application. The remaining 30 rats was divided into two equal diazinon groups (n=10) and the control one (corn oil). The first group treated during 7 days, and second during 14 days with 55 mg/kg of diazinon (1/10 of previously determined LD 50 value). The histopathology of the pancreas and liver and relative activities of LDH isoenzymes in the blood were determined after the completion of both time period.Results. Single administration of increasing doses of diazinon, results in a significant increase of concentrations of glucose, activity of α-amylase and LDH isoenzymes.Subacute application of low diazinon dose induces histopathological changes in the pancreas manifested by acinar cell necrosis, and in the liver in the form of portal hepatitis and multifocal necrosis. The cumulative doses resulted in statistically significantly lower activities of LDH isoenzymes compared with the single administration of these doses, indicating a lower degree of the cells damage after subacute diazinon administration. Conclusion.Subacute administration of low dose of diazinon leads to the different degree adaptation of the organs and organ systems to toxic effects caused by this organophosphate.
