Passive IgE‐sensitization by blood transfusion

Abstract: Transfused IgE antibodies will sensitize mast cells and basophils to CD-sens levels similar to those of allergic patients. The recipients expressed 'slow' or 'rapid' CD-sens decline, indicating two different basophil populations. After transfusion of plasma with >10 kU(A)/l IgE antibody the recipient could have allergen reactive basophils for up to 7 weeks.

“…The observation that basophils passively sensitized in vivo by low amounts specific IgE respond to the appropriate allergen within up to 2 weeks supports this idea. 54 Our data show that spontaneous apoptosis of basophils operates through a caspase-dependent mechanism. It is associated with catalytic processing of caspase 3 into its active form (p17/p12 dimer) and the subsequent cleavage of caspase 3 intracellular substrates, PARP and Bcl-2.…”

“…The same group of investigators subsequently performed an in vivo study in which two units (each approximately 300 ml) containing known concentrations of IgE antibodies to timothy grass pollen (8-205 kilo units antigen (kUA)/l) were selected and transfused into patients (Johansson et al, 2005b). IgE in the transfused plasma could be detected in the circulation of the recipients within 3 h after transfusion; the mean half-life of IgE was 1Á13 d. Basophil sensitization was obvious in the 3 h sample, and it increased rapidly to a peak after 3Á4 d and then declined over days to weeks.…”
