Controlled infection systems constitute a valuable tool in the study of pathosystems, as they enable the observation and measurement of specific characteristics with greater precision and ease compared to natural conditions. Additionally, they are useful for exploring and identifying sources of resistance in host populations. However, the plant's response to infection can vary depending on the inoculation method; invasive methods can damage plant parts, thus facilitating pathogen entry even in the presence of certain resistance mechanisms. The purpose of this study was to evaluate the reproducibility of an artificial infection protocol and an assessment scale for primary symptoms at the stem base/root collar, as well as secondary symptoms in leaves and roots of two Passiflora species. We evaluated the effects of inoculating planting substrates of sweet granadilla and purple passion fruit plants with three concentrations of F. solani f. sp. passiflorae (0.5 × 106, 1 × 106, and 2 × 106 conidia gram-1 of substrate), using non-inoculated plants as controls under controlled conditions. These treatments were tested in two separate experiments performed in 2017 and 2020. The study included symptom development readings integrated in a disease severity index (SI) for collar rot. Both experiments showed severity indices greater that 56%, indicating a high level of efficacy of the inoculation system for replicating disease symptoms in greenhouse settings. Our inoculation method reflects the conditions of natural infection of F. solani in sweet granadilla and purple passion fruit plants; it could be suitable for epidemiological and disease control studies, as well as for routine germplasm screening to identify sources of resistance.

The inhibitory effects and mechanisms of eugenol were studied in pathogenic fungi that were isolated from passion fruit affected with naturally occurring fruit rot; additionally, the effect of eugenol treatment on fruit rot resistance in passion fruit was investigated. The corresponding results demonstrated that the pathogenic fungus causing passion fruit rot was Lasiodiplodia theobromae and that the minimal inhibitory concentration of eugenol against L. theobromae was 0.30 mg/mL. Eugenol significantly inhibited mycelial growth and spore germination of this fruit rot fungus. Further, nucleic acid release, electrical conductivity, and protein and soluble sugar content of the fruit rot fungus gradually increased with increasing eugenol concentrations. Propidium iodide staining revealed that the cell membrane integrity of L. theobromae hyphae was disrupted when treated with eugenol. In addition, eugenol treatment inhibited the spread of disease spot diameter after inoculation with L. theobromae, effectively increased APX, SOD, CAT, POD, 4CL, C4H, and PAL activities, promoted the accumulation of disease-stage-related proteins CHI and GLU, and increased the total phenol and flavonoid content during storage in passion fruit. Overall, these results suggest that eugenol has good application prospects for the effective control of fruit rot in passion fruit.

Passion fruit is widely cultivated in tropical, subtropical regions of the world. The attack of bacterial and fungal diseases, and environmental factors heavily affect the yield and productivity of the passion fruit. The CC-NBS-LRR (CNL) gene family being a subclass of R-genes protects the plant against the attack of pathogens and plays a major role in effector-triggered immunity (ETI). However, no information is available regarding this gene family in passion fruit. To address the underlying problem a total of 25 and 21 CNL genes have been identified in the genome of purple (Passiflora edulis Sims.) and yellow (Passiflora edulis f. flavicarpa) passion fruit respectively. Phylogenetic tree was divided into four groups with PeCNLs present in 3 groups only. Gene structure analysis revealed that number of exons ranged from 1 to 9 with 1 being most common. Most of the PeCNL genes were clustered at the chromosome 3 and underwent strong purifying selection, expanded through segmental (17 gene pairs) and tandem duplications (17 gene pairs). PeCNL genes contained cis-elements involved in plant growth, hormones, and stress response. Transcriptome data indicated that PeCNL3, PeCNL13, and PeCNL14 were found to be differentially expressed under Cucumber mosaic virus and cold stress. Three genes were validated to be multi-stress responsive by applying Random Forest model of machine learning. To comprehend the biological functions of PeCNL proteins, their 3D structure and gene ontology (GO) enrichment analysis were done. Our research analyzed the CNL gene family in passion fruit to understand stress regulation and improve resilience. This study lays the groundwork for future investigations aimed at enhancing the genetic composition of passion fruit to ensure robust growth and productivity in challenging environments.
