Parvalbumin-containing interneurons do not innervate granule cells in the olfactory bulb

Abstract: Combining pre-embedding parvalbumin immunostaining and post-embedding immunogold detection of GABA in the olfactory bulb, we investigated whether the parvalbumin-containing GABAergic interneurons of the external plexiform layer exclusively innervate principal cells, or whether they also establish inhibitory synapses upon GABAergic local neurons such as granule cells. Our results demonstrate that the parvalbumin-containing cells do not contact GABAergic interneurons in the neuropil of the external plexiform lay… Show more

“…Regarding the putative functional role of Van Gehuchten cells in MOB circuitry, serial electron microscopy studies showed that VIP and/or PV processes directly innervate M/T cells (Toida et al, 1996;Crespo et al, 2001Crespo et al, , 2002. This challenges the classical scheme according to which EPL interneurons only contact granule cells (Schneider and Macrides, 1978).…”

“…This is mediated largely through the actions of a diverse set of interneurons (Aungst et al, 2003;Lledo et al, 2005). Subpopulations of these interneurons can be characterized by their location, synaptic connectivity, morphology, firing patterns, and expression of a variety of immunomarkers (Price and Powell, 1970a,c;Pinching and Powell, 1971a;Shipley and Ennis, 1996;Crespo et al, 2001;McQuiston and Katz, 2001;Kosaka and Kosaka, 2005).…”

“…In rat olfactory bulb, for example, immunoreactivity for non-phosphorylated neurofi lament protein labels mitral cells and tufted cells [Paxinos et al, 1999]. In several eutherian species, immunohistochemistry to the three calcium binding proteins identifi es a variety of neuron types [Briñón et al, 1992[Briñón et al, , 1997[Briñón et al, , 1999[Briñón et al, , 2001Kosaka et al, 1994;Toida et al, 1996;Alonso et al, 2001;Crespo et al, 2001;Jia and Halpern, 2004]. Immunoreactivity for tyrosine hydroxylase is intense within periglomerular cells of many eutherian species [Baker, 1986a,b;Smith et al, 1991;Brunjes et al, 1992], whereas NADPH diaphorase reactive cells have been found in superfi cial and deep short axon cells of some eutherian [Scott et al, 1987].…”

“…identifi es a wide range of cell types, including periglomerular cells, superfi cial and deep short axon cells, van Gehuchten cells, multipolar cells and horizontal cells [Kosaka et al, 1994;Toida et al, 1996;Crespo et al, 2001]. By contrast, PV immunoreactivity identifi es fewer cells in the olfactory bulb of certain primates with only a few superfi cial and deep short axon cells and some small interneurons in the EPl being labeled .…”
