Partitioning of cerrena unicolor laccase activity in an aqueous two-phase system

Abstract: Culture supernatant containing laccase produced by Cerrena unicolor strain was used to examine laccase partitioning between phases in an aqueous two-phase system. The investigated system consisted of polyethylene glycol 3000 and sodium phosphate buffer adjusted to pH = 7. Influence of several parameters on partitioning was measured, including phase forming components' concentrations, tie line lengths, phase volume ratio, supernatant dilution, process temperature and halogen salt supplementation. Partitioning c… Show more

“…18, 22, 25) gave relatively high concentration factors because the majority of the enzyme was concentrated within a small volume. Both phenomena were described in the previous work (Blatkiewicz et al 2016 ). It can also be noticed that such points were encumbered with relatively high measurement errors.…”

“…As proven in the previous work (Blatkiewicz et al 2016 ), the difference between the volumes of separated phases is a very important parameter for ATPE. Therefore, the volume fraction (VF) is also considered as one of the key parameters in this work.…”

“…Therefore, a lot of research is focused on finding and developing novel, cost-effective, efficient methods of bioproduct separation and recovery with satisfying selectivity towards the desired product and granting its stability in the process. Various downstream processing methods of laccase have been investigated, including chromatographic methods (Scherer and Fischer, 1998) and aqueous two-phase extraction (Blatkiewicz et al, 2016;Prinz et al, 2014). One of the promising, currently investigated methods is foam fractionation (FF).…”
