Particulate guanylyl cyclases: multiple mechanisms of activation.

Abstract: Cyclic GMP (cGMP), a key messenger in several signal transduction pathways, is synthesized from GTP by a family of enzymes termed guanylyl cyclases, which are found in two forms: cytosolic (soluble) and membrane-bound (particulate). The past decade has brought significant progress in understanding the molecular mechanisms that underlie the regulation of particulate guanylyl cyclases and new members of their family have been identified. It has become more evident that the basic mechanism of catalysis of guanyly… Show more

“…In the mammalian retina, three GCAPs (GCAP1, GCAP2 and GCAP3) have been identified Palczewski et al, 1994;Dizhoor et al, 1995;Haeseleer et al, 1999;Kobialka & Gorczyca, 2000;Imanishi et al, 2002;Gorczyca et al, 2003) that regulate the activity of photoreceptor GCs in Ca 2+ -dependent manners. GCAPs belong to the family of recoverin-like proteins with limited homology to CaM, and they are myristoylated at the N-terminus .…”

“…Given the impaired sGC activity, this normalization may involve membranebound or particulate guanylyl cyclase (pGC). The activity of pGC can be stimulated by natriuretic peptides (ANP and BNP) (Kobialka and Gorczyca 2000). It is The Physiological Society and the American Physiological Society possible that the significant increase in PAP during incremental exercise causes secretion of natriuretic peptides by cardiomyocytes in response to cardiac stretch (Wong et al 2017), and thus pGC activation.…”

“…1 A first-in-human clinical trials 5 carried out demonstrated that subjects infused with CD-NP at 17.5 ng/(kg min) for 4 h had significant plasma and urinary 3 ,5 cyclic guanosine monophospate (cGMP) levels compared against the placebo group. Significant elevation of cGMP is believed to be attributed to CD-NP's dual activation of GC-A and GC-B, [14][15][16] whereas native NPs can only activate one of GC receptors, which is either GC-A 3,12,14 or GC-B. [17][18][19] Furthermore, the urinary sodium excretion and urine flow in CD-NP treatment groups were found to be significant compared with placebo, which indicated that CD-NP achieved overall favorable cGMP-stimulation, natriuretic responses, and absence of excessive hypotension in healthy adults, further boosting CD-NP's clinical usage.…”
