APMIS 96: 660-666, 1988.Particulate iodipamide ethyl ester, a new hepatolienographic x-ray contrast agent, was intravenously injected into rats. Lung and kidney biopsies taken at various intervals after the injection were examined by light and electron microscopy. IDE particles could be found in the lung capillaries phagocytized by polymorphonuclear neutrophils (PMNs). There were also free particles in the alveolar capillaries in the samples taken 5 min to 4 hours after the injection. No aggregates or emboli were seen. Two days or more after the injection no intra-and extracellular particles were present. The PMNs underwent transient local hydropic degeneration; the lung cells were morphologically intact. In the kidneys, the particles first appeared in both cortical and medullary capillaries. No emboli were observed. The kidney cells did not ingest IDE, but polymorphonuclear neutrophils (PMNs) with ingested IDE were often seen loosely attached to the glomerular capillary walls. In addition, free particles were evident in the capillaries in the samples taken up to 1 hour after injection. All particles in subsequent kidney samples were located in PMNs in the glomeruli. After three or more days the renal tissue was totally devoid of particulate IDE. No morphological evidence of kidney cell injury was observed.

We have compared responses in platelets stimulated with the particulate materials, Intralipid (liposome-suspension) and a potential contrast medium IEEC (1P-(ethyloxycarbonyloxy)-ethyl-5-acetyl-amino-3-(N-methyl-acetylamino)-2,4,6-triiodo-benzenecarboxylate coated with human serum albumin), with and without forskolin and inhibitors of autocrine stimulation (IAS: an ADP-removing system of creatine phosphate/creatine phosphokinase; RGDS to prevent fibrinogen/fibronectin binding to GPIIb/IIIa ; SQ 29.548 as a TXA P receptor antagonist; cyproheptadine as a serotonin receptor antagonist; BN 52021 as a platelet-activating factor receptor antagonist). The pattern of tyrosine-phosphorylated proteins, phosphorylation of initial lipids and phosphorylation of pleckstrin (P47) were used as markers for early signal transducing responses, while secretion of ADP+ATP and L L-N-acetyl-glycosaminidase were used as final responses. Intralipid showed no platelet activation except for some weak tyrosine protein phosphorylation that was inhibited by elevated cAMP. IEEC induced strong platelet activation that was partly inhibited by increased levels of cAMP and IAS. The inhibition of elevated cAMP seemed to be due to removal of the G protein-mediated activation from secreted autocrine stimulators either by IAS or forskolin. The remaining activity is a pure effect from IEEC which is not affected by elevated cAMP.z 1998 Federation of European Biochemical Societies.

This review discusses radiopaques from research and development to application. Radiopaques attenuate X‐rays and may be either high atomic‐numbered materials or iodinated organic compounds. The latter may appear sometimes in liposomes or particulates. The theoretical and historical aspects, classification by structures, methods of synthesis, structure‐activity relationship, physical, chemical, and pharmacological properties, and related reactions, essential to the understanding of safety, tolerability, and efficacy of radiopaques in medical use, are included in the discussion.
