Particle tracking microrheology of purified gastrointestinal mucins

Abstract: ABSTRACT:The rheological characteristics of gastric and duodenal mucin solutions, the building blocks of the mucus layer that covers the epithelia of the two organs, were investigated using particle tracking microrheology. We used biochemically well characterized purified porcine mucins for MUC5AC and h $ c 5:160:8 for MUC2. Studies by Georgiades et al have shown that GuHCl disassembles the semi dilute network structure of porcine gastric mucin solutions by unfolding proteins and disrupting non-covalent interactions [17]. In studies where it is necessary to visualize particles in a scraped mucus gel or the gel contains food particles and other debris, the gel can be cleaned up by gently stirring for one hour at 4 °C in sodium chloride (0.1M) (1:5 w/v mucus to sodium chloride) and then centrifuged (10,400g at 10 °C) for 2 hours.…”
