Participation of Serotonin-Containing Neurons in the Suckling-Induced Rise in Plasma Prolactin Levels in Lactating Rats

Abstract: Plasma prolactin levels were measured by radioimmuno-assay in free-moving lactating rats bearing heart cannulas. Litters adjusted to 6 pups were returned to the mothers after a separation of 8–12 h from days 6 to 15 postpartum. Blood samples were taken before and 15, 30, 60 and 120 min after suckling had resumed. Lactating mothers were treated at various intervals before the experiment with P-chlorophenylalanine (PCPA), an inhibitor of serotonin (5-HT) synthesis; some animals were treated in addition with 5-hy… Show more

“…In animal studies, inhibition of both serotonin 43 and endogenous opioids 44 blocks suckling-induced prolactin release and milk transfer. Prolonged stress appears to decrease the effect of endogenous opioids on prolactin secretion, 45 suggesting that tonic hypothalamic-pituitary-adrenal (HPA) axis activation may inhibit prolactin production.…”

“…Conversely, blocking either serotonin synthesis or transmission results in a blunted PRL release (Kordon et al, 1973;Gil-ad et al, 1976;Kamberi et al, 1971). These effects depend on one or more PRFs, making serotonin an indirect, albeit potent, modulator of PRL release (BenJonathan, 1994;Freeman et al, 2000) (Figure 1).…”

“…In this paper we present the results of our investigations on the nature and the regulation of prolactin release after stimulation of lactating ewe mammary gland. Most recent experiments on prolactin regulation during nursing have been performed on the polytocous female rat Kordon et al, 1973 ;Mena et al, 1976). We propose that the ewe might be a model for females having one or two young at each parturition.…”
