Partial rescue of postnatal growth plate abnormalities in Ihh mutants by expression of a constitutively active PTH/PTHrP receptor

Abstract: Indian hedgehog (Ihh) is essential for chondrocyte proliferation/differentiation and osteoblast differentiation during prenatal endochondral bone formation. Ihh expression in postnatal chondrocytes has a non-redundant role in maintaining a growth plate and sustaining trabecular bone after birth. Loss of Ihh in postnatal chondrocytes results in fusion of the growth plate and a decrease in trabecular bone. In order to normalize this abnormal chondrocyte phenotype and to investigate whether a putative rescue of t… Show more

“…Ihh is a critical factor secreted by chondrocytes, but its expression is restricted mainly to the hypertrophic zone (14). It is well known that increased activity of the Ihh signaling pathway promotes chondrocyte proliferation (57) and mutation or disruption of Ihh signaling results in reduction of the height of the columnar layer in the growth plate (58). GCs are also known to suppresses Hh activity in neurons (59).…”

“…6,7). To study this disease in more detail, a transgenic mouse model expressing the H223R mutation under the control of the collagen ␣1(II) promoter, the Jansen mouse, has been generated (7,10,11). Interestingly, serum parathyroid hormone (PTH) levels are normal to low, distinguishing it from hyperparathyroidism (8,9).…”

To study this disease in more detail, a transgenic mouse model expressing the H223R mutation under the control of the collagen ␣1(II) promoter, the Jansen mouse, has been generated (7,10,11). In both mice and humans, the mutant PTHR1 leads to cAMP accumulation, and thereby constitutive signaling, despite the absence of a ligand.…”

“…In this study, a conditional Ihh knockout mouse was generated that expressed a constitutively active PPR specifically in chondrocytes (Maeda et al, 2009). The authors showed that expression of the PPR was able to transiently maintain a growth plate.…”
