Partial Purification of the Cyanide-Resistant Alternative Oxidase of Skunk Cabbage (Symplocarpus foetidus) Mitochondria

Abstract: A partial purification of the cyanide-resistant, alternative oxidase from skunk cabbage (Symplocarpus foetidus L.) spadix mitochondria is described. Skunk cabbage mitochondria were solubilized in N,N-bis-(3-~-glucon-amido-propyl)deoxycholamide and the alternative oxidase was purified using a batch DEAE-cellulose treatment, followed by precipitation with Extracti-Cel and chromatography on Sephadex C-200. Following pooling and concentrating of the most active fractions from the gel filtration column, a 20-to 30-… Show more

“…For AOX, the large g-anisotropy (see Davydov et al,Ref. 33), the small value of the exchange coupling constant (ϪJ), the fact that the EPR signal is seen only at very low temperature, and the lack of a visible absorption at wavelengths above 320 nm (10), all point to the presence of a hydroxo (rather than oxo) bridge between the two irons. In fact, the parameters also leave open the possibility that the bridging ligand may be an aquo group.…”

“…Careful attempts made earlier to locate an EPR signal in the enzyme in whole mitochondria had been unsuccessful (35)(36)(37), as were EPR analyses of partially purified preparations (10,38).…”

“…A partially purified alternative oxidase preparation from Symplocarpus foetidus spadices also showed no EPR resonances in either the oxidized or the dithionitereduced states and had no optical absorbance above 350 nm [17]. Preliminary analysis of one S. foetidus preparation using atomic absorption spectroscopy has indicated the presence of iron (A.L.…”

“…The current model of the AOX, supported by mutagenesis studies, predicts a monotopic integral membrane protein (2,(13)(14)(15) associating with one leaflet of the lipid bilayer. Although analyses of yeast and trypanosomal enzymes have established that iron is required for activity (16,17), early investigations of either mitochondria or partially purified protein failed to reveal spectroscopic signatures of its active site (18,19). The first spectroscopic evidence for iron involvement was provided by Berthold et al (20) (20) were not successful.…”
