Partial characterization of bacteriocin-like substance produced by probiotic Lactobacillus plantarum F12 isolated from Algerian children faeces

Abstract: The strain, Lactobacillus plantarum F12 with probiotic traits was isolated from Algerian healthy children faeces and identified by 16S rDNA sequencing. In this study, the antimicrobial activity and physicochemical properties of bacteriocin-like substance (BLS) produced by this strain were determined. Also, the bacteriocinogenic genes of plantaricin A, plantaricin J and plantaricin K were screened in this strain. The BLS inhibited a range of pathogenic and spoilage bacteria including Escherichia coli, Salmonell… Show more

“…The cell-free supernatant was subjected to ammonium sulfate fractionation. On centrifugation at 10,000 rpm and temperature of 4 °C for 10 min, the pellets were collected and resuspended in a minimal volume of 0.2 ml Tris-HCl buffer pH 7.0 (Karthikeyan and Santosh 2009). A dialysis tube was treated to remove protectants such as sulfur or glycerin compounds present in it.…”

“…Purified pediocin has also been characterized to test its stability against temperature, pH, and proteolytic enzymes. According to Karthikeyan and Santosh [6], purified pediocin SM-1 was stable for 60 min at 100 o C, even at 121 o C. Storage for one year at a temperature of -80 o C and -20 o C did not affect its activity. SM-1 pediocin resistance is in the pH 3.0-12.0 and inactive at pH 2.0, 13.0, and 14.0.…”

“…Bacteriocins are antimicrobial peptide compounds derived from gram-positive and gram-negative bacteria. Bacteriocins can be cationic, anionic, and neutral [6]. These compounds are synthesized in bacterial ribosomes and have varied activities and a broad antimicrobial spectrum.…”

“…The use of LAB was the safest microorganism in the development of probiotics for aquaculture due to their antagonistic activity against opportunistic pathogens, fungi, and viruses that cause microbiological spoilage of feed, pollute water bodies, and cause diseases in aquatic animals (Chizhayeva et al 2022). Karthikeyan and Santosh (2009) isolated Lactobacillus plantarum from the intestines of black tiger shrimp (Penaeus monodon Fabricius, 1798) with the ability to produce bacteriocin against some pathogenic bacteria such as Salmonella typhimurium, Vibrio cholera, and Staphylococcus aureus. On the other hand, LAB strains were also isolated from the fish intestine, and some fermentation products were inhibitory to pathogenic bacteria in seawater and freshwater aquatic animals (Alonso et al 2018;Lim et al 2020;.…”
