PARP1 activation increases expression of modified tumor suppressors and pathways underlying development of aggressive hepatoblastoma

Abstract: Hepatoblastoma (HBL) is a pediatric liver cancer that affects children under the age of three. Reduction of tumor suppressor proteins (TSPs) is commonly seen in liver cancer. However, in our studies we find that aggressive, chemo-resistant HBLs exhibit an elevation of TSPs. HBL patients with a classic phenotype have reduced TSP levels, but patients with aggressive HBL express elevated TSPs that undergo posttranslational modifications, eliminating their tumor suppression activities. Here we identify unique aggr… Show more

“…Protein extracts were isolated from available 10 HBL and 5 NTCL samples as previously described. 27,28 This assay was performed in duplicate, where proteins (50 µg) were loaded onto a 4% to 20% gradient gel (BioRad, USA) and transferred to a nitrocellulose membrane (BioRad). The membranes were probed with anti-NNMT (OTI3D8; Abcam) and anti-beta-actin (ab8227; Abcam) antibodies.…”

“…Increased expression of EPHA8 may be a prognostic marker for epithelial ovarian cancer [28]. The effect of increased expression of PARP1 in HCC has also been previously reported [29,30]. PARP1 has been shown to physically interact with hepatitis B virus X protein and to inhibit the activity of the DNA repair complex at damaged DNA sites, resulting in hepatocarcinogenesis [31].…”

“…PARPs are multifunctional enzymes involved in epigenetic modifications, signal transduction, stress sensing, and DNA repair ( 60 62 ). Interestingly, in a context of aggressive HB, PARP1 was recently shown to be aberrantly activated, promoting expression of nonfunctional tumor suppressor proteins ( 63 ). Herein, we demonstrated decreases in PARP1 and PARP2 expression in HB cells both at mRNA and protein level after CQ treatment.…”
