Parameter optimization for the enzymatic hydrolysis of sunflower oil in high‐pressure reactors

Abstract: This study is concerned with the hydrolysis of sunflower oil in the presence of lipase preparation Lipolase 100T (Aspergillus niger lipase). Supercritical carbon dioxide was used as a solvent for this reaction. In a high-pressure stirred tank reactor operated in a batch mode, the effects of various process parameters (temperature, pressure, enzyme/substrate ratio, pH, and oil/buffer ratio) were investigated to determine the optimal reaction rate and conversion for the hydrolysis process. The optimal concentrat… Show more

“…The moisture content of the bound lipase was about 3.85% after 48 h of freeze drying. The presence of a small amount of bound water or biocatalyst water in an enzyme is crucial for the activation of biocatalyst at the oil-water interface (Zaks and Klibanov 1988;Primozic et al 2003). If this water is stripped away, the activity of the enzyme falls (Primozic et al 2003).…”

“…The presence of a small amount of bound water or biocatalyst water in an enzyme is crucial for the activation of biocatalyst at the oil-water interface (Zaks and Klibanov 1988;Primozic et al 2003). If this water is stripped away, the activity of the enzyme falls (Primozic et al 2003). During vacuum drying of the mycelia cell, rapid freezing is important for the stability of the enzyme to be lyophilized since low freezing may result in ''freezing out'' of solution components and some bound water may be stripped from the MBL, leading to concentration gradients and substantial shifts of pH that could cause the denaturation of the protein.…”

“…The mechanisms and the kinetics involved in triacylglycerol (TAG) hydrolysis to produce fatty acids from vegetable oils, including soybean oil, by the action of various lipases, have been studied extensively (Freitas, Bueno, Perez, Santos, & de Castro, 2007;Liu et al, 2008;Primozic, Habulin, & Knez, 2003;Ramachandran, AlZuhair, Fong, & Gak, 2006;Ting, Tung, Giridhar, & Wu, 2006). The intent has been to optimise the hydrolytic conditions and identify possible enhancing technologies to increase the yield of fatty acids.…”

“…This may explain why a flow rate for substrates of 0.1 mL/min resulted in a higher extent of lipolysis than higher flow rates. A greater degree of hydrolysis of sunflower oil by Lipolase 100T was achieved using a high-pressure batch stirred tank reactor, but not until after 48 h of operation (3). With the HP CFSMR, steady-state lipolysis was generally achieved within 1 h and subsequently maintained.…”

“…Optimal parameters for the hydrolysis of sunflower oil in a high-pressure batch stirred tank reactor (3). Optimal reaction parameters, determined previously for the determination of thermodynamic parameters, included optimal lipase concentration, 0.0714 g/mL of CO 2 -free reaction mixture; optimal pressure, 200 bar; optimal buffer/oil ratio, 1:1 (w/w); and optimal temperature, 50°C.…”
