Parafibromin governs cell polarity and centrosome assembly in Drosophila neural stem cells

Abstract: Neural stem cells (NSCs) divide asymmetrically to balance their self-renewal and differentiation, an imbalance in which can lead to NSC overgrowth and tumor formation. The functions of Parafibromin, a conserved tumor suppressor, in the nervous system are not established. Here, we demonstrate that Drosophila Parafibromin/Hyrax (Hyx) inhibits ectopic NSC formation by governing cell polarity. Hyx is essential for the asymmetric distribution and/or maintenance of polarity proteins. hyx depletion results in the sym… Show more

“…Super-resolution imaging was performed as previously described [60]. In brief, super-resolution Spinning Disc Confocal-Structured Illumination Microscopy (SDC-SIM) was performed on a spinning disk system (Gataca Systems) based on an inverted microscope (Nikon Ti2-E; Nikon) equipped with a confocal spinning head (CSU-W; Yokogawa), a Plan-Apo objective (100×1.45-NA), a back-illuminated sCMOS camera (Prime95B; Teledyne Photometrics) and a super-resolution module (Live-SR; GATACA Systems).…”
