It is well known that buffaloes have seasonal anoestrus and that the use of melatonin reduces the effects of season in reproduction, articularly among sheep and goats. This study evaluates the use of melatonin on water buffaloes to increase pregnancy rates in a fixed-time insemination programme. The study was conducted on an Argentinian buffalo herd, located in the Corrientes Province, during an unfavourable reproductive season. Artificial insemination cycles were performed in September and December 2018. Sixty animals were selected, and melatonin was administered to 29 females at a dose of 18 mg/50 kg. Ten days after the melatonin implant, ovulation synchronization was started. Animals were inseminated with frozen semen of a single bull. Pregnancy was evaluated by ultrasound 35 days after insemination. The pregnancy rate of the two repetitions was 34.62% for September and 6.4% for December (P=0.007). No significant differences were found in the parameters associated with follicular development and formation of the corpus luteum. Analysis of the effects of melatonin on the two replicates showed that there was no statistically significant effect of ovarian response regarding the analysed follicular development parameters. There was also no effect on pregnancy rates (17.85% vs. 20.68%; P=0.15), or on follicular development for treatment and control. In conclusion, this study was not able to demonstrate an effect of melatonin on the ovarian response of buffaloes as reported by other authors. Other factors, such as environmental conditions of subtropical areas and species specificities, may have a more significant effect on buffalo endocrinology. These factors should be evaluated to improve the results of fixed-timed artificial insemination programmes.

Impact of in vitro fertilization by refrigerated versus frozen buffalo semen on developmental competence of buffalo embryos

The objective of this study was to evaluate the fertility of buffalo semen for in vitro embryo production (IVEP) by comparing the effectiveness of refrigerated versus frozen semen. Three OPU sessions were held at 30-day intervals. For oocyte fertilization three buffalo bulls were used, one per session. At each OPU-IVEP session, one ejaculate was collected and divided into two equal aliquots. Each aliquot was either refrigerated at 5ºC/24 hours or frozen. A TRIS extender containing 10% low density lipoproteins, 0.5% lecithin and 10 mM acetylcysteine was used adding 7% glycerol for freezing. Sperm motility/kinetic was evaluated by CASA and sperm membrane integrity by the hypoosmotic swelling test. The evaluations were performed at 0 h (post final dilution at 37ºC), at 4 and 24 hs post-incubation at 5ºC and post-thaw. At 24 hs incubation and immediately post thaw sperm cells were used for in vitro fertilization of buffalo oocytes equally distributed between both groups. Cleavage rates and embryo development were followed. The embryo/matured and embryo/cultured rates were 25.4 x 14.0% and 29.4 x 18.5% (P<0.05), for chilled and frozen semen, respectively. It is concluded that cooled semen can be used for in vitro embryo production in buffalo and that a better efficiency may be expected for cooled compared to frozen semen.

A criação de búfalos vem se difundindo mundialmente devido à superioridade que pode apresentar em relação aos outros ruminantes domésticos, principalmente a adaptação às diversas condições climáticas do território brasileiro, sendo que ele poderá produzir carne e leite para suprir as necessidades do mercado nacional, propiciando grande contribuição ao atendimento da demanda familiar. A exploração zootécnica dos bubalinos caracteriza-se por apresentar boa eficiência reprodutiva e rápido desenvolvimento ponderal, sendo que este potencial genético pode ser multiplicado mais rapidamente por biotécnicas da reprodução, pois os mercados nacional e internacional de bubalinos necessitam de animais geneticamente superiores. Portanto, devido à importância da criação de bubalinos para o País, o objetivo desta revisão bibliográfica é abordar alguns aspectos da fisiologia reprodutiva e a utilização de biotécnicas reprodutivas nesta espécie, a fim de se promover melhorias para a seleção zootécnica e o desempenho destes animais.Palavras chave: inseminação artificial, melhoramento genético, produtividade, transferência de embriões Reproductive Biotechniques applied to buffalos (Bubalus Bubalis)ABSTRACT. The livestock use of buffaloes is characterized by the good reproductive efficiency and fast body weight gain, and this genetic potential can be multiplied faster by reproductive biotechniques, Buffalo's national and international business markets requires superior genetically animals. Therefore, due to the importance of buffalo breeding for the country, the aim of this review is to describe some aspects of reproductive physiology and the use of reproductive biotechniques in this species, to promote improvements to the genetic selection and performance of these animals.
