Paradoxical antidepressant effects of alcohol are related to acid sphingomyelinase and its control of sphingolipid homeostasis

Abstract: Alcohol is a widely consumed drug that can lead to addiction and severe brain damage. However, alcohol is also used as self-medication for psychiatric problems, such as depression, frequently resulting in depression-alcoholism comorbidity. Here, we identify the first molecular mechanism for alcohol use with the goal to self-medicate and ameliorate the behavioral symptoms of a genetically induced innate depression. An induced over-expression of acid sphingomyelinase (ASM), as was observed in depressed patients,… Show more

“…These findings provide the first mechanistic evidence for alcohol instrumentalization with the goal to self-medicate and ameliorate behavioral symptoms of a genetically induced innate depression. From that, sphingolipid homeostasis emerged as a new mechanism to control depression-alcohol addiction comorbidity (Müller et al 2016 ).…”

“…SMPD4 is also highly expressed in the brain and is important in maintaining sphingolipid metabolism in neuronal system. Sphingolipid homeostasis has been associated with the effects of alcohol on depression [22]. ASE analysis examines the transcriptional differences between two alleles and is designed to identify genes whose expression levels are influenced by the cis-acting elements.…”
