Paracrine Insulin-Like Growth Factor Signaling Influences Primordial Germ Cell Migration: In Vivo Evidence from the Zebrafish Model

Abstract: IGF signaling has been shown to stimulate migration of multiple cell types in vitro, but few studies have confirmed an equivalent function for IGF signaling in vivo. We recently showed that suppression of IGF receptors in the zebrafish embryo disrupts primordial germ cell (PGC) migration, but the mechanism underlying these effects has not been elucidated. We hypothesized that PGCs are intrinsically dependent upon IGF signaling during the migratory phase of development. To test this hypothesis, we first examine… Show more

“…IGF-2a and -IGF-2b amino acid sequences share 65 % identity (data not shown). Both genes are maternally deposited and dynamically expressed during embryonic development (Maures et al, 2002;Eivers et al, 2004;Sang et al, 2008). In addition, we show igf-2b expression in the embryonic shield, in anterior neural tissues and the developing heart ( Supplementary Fig.…”

“…IGF-2 is more highly expressed than IGF-1 during development, indicating that IGF-2 may be the more important IGF ligand during embryonic development (Sang et al, 2008). Two igf-2 genes are present in the zebrafish genome, igf-2a and igf-2b.…”

“…SLa and SLb could act on growth development to some extent as GH does -through IGF signaling. Recently, IGFs have been found to be expressed in teleosts in a wide range of tissues throughout life, which suggests that the autocrine/paracrine role of this hormone may be of particular physiological importance in bony fish (Schlueter et al 2007, Sang et al 2008, Wang et al 2008. For example, gonad-specific IGF3 was found to be present only in the fish genome (Sang et al 2008), which suggests its potential function in gonad development and N o n -t r a n s g e n i c S L 伪 -t r a n s g e n i c S L 尾 -t r a n s g e n i c G H -t r a n s g e n i c 1路45 a ** P < 0路01 * P < 0路05 a Relative POMC mRNA level N o n -t r a n s g e n i c S L 伪 -t r a n s g e n i c S L 尾 -t r a n s g e n i c G H -t r a n s g e n i c N o n -t r a n s g e n i c S L 伪 -t r a n s g e n i c S L 尾 -t r a n s g e n i c G H -t r a n s g e n i c Different letters indicate significant differences (P!0 .…”

“…IGF2 is more highly expressed than IGF1 during development in rodents, ruminants and humans (Delhanty & Han 1993, Hill & Pell 1998, suggesting that it may be the more important IGF during development (Sang et al 2008). Recently, Hills et al (2012) have described IGF2 as a more effective anti-apoptotic survival factor compared with IGF1 in human placental villous cytotrophoblasts.…”
