Pantetheinase activity of membrane‐bound Vanin‐1: lack of free cysteamine in tissues of Vanin‐1 deficient mice

Abstract: Pantetheinase (EC 3.5.1.-) is an ubiquitous enzyme which in vitro has been shown to recycle pantothenic acid (vitamin B5) and to produce cysteamine, a potent anti-oxidant. We show that the Vanin-1 gene encodes pantetheinase widely expressed in mouse tissues : (1) a pantetheinase activity is specifically expressed by Vanin-1 transfectants and is immunodepleted by specific antibodies; (2) Vanin-1 is a GPI-anchored pantetheinase, and consequently an ectoenzyme; (3) Vanin-1 null mice are deficient in membrane-boun… Show more

“…Cysteamine promotes the production of chemokines that attract neutrophils and inhibits ␥-glutamylcysteine synthetase, which is needed to synthesize the natural reducing agent glutathione (69,70). This suggests that increased Vanin-1 is likely to exacerbate inflammation dependent on neutrophils and oxidation, which is consistent with the decreased inflammation observed in S. mansoni-infected, Vnn1-deficient mice (71).…”

“…Vanin-1 genotyping was done by PCR. 26 The ank/ank/Vnn1 Ϫ/Ϫ mice were generated by crossing Ank/ank and Vnn1 Ϫ/Ϫ mice to generate double heterozygotes which were bred to generate Ank/Ank/ Vnn1 ϩ/ϩ , Ank/Ank/Vnn1 Ϫ/Ϫ , ank/ank/Vnn1 ϩ/ϩ , and ank/ ank/Vnn1 Ϫ/Ϫ littermates.…”

“…9,22,23,25 Pantetheinases specifically hydrolyze pantetheine to pantothenic acid (vitamin B 5 ) and the cell-permeant sulfhydryl cysteamine (NH 2 -CH 2 -CH 2 -SH). 26,27 Cysteamine directly inhibits ␥-glutamylcysteine synthetase, the rate-limiting enzyme in synthesis of GSH. 22,23 Vnn1 Ϫ/Ϫ mice, which have a grossly normal phenotype, lack free cysteamine in tissues and demonstrate elevated stores of GSH in multiple tissues.…”

“…Vanin-1-deficient mice develop normally but their tissues lack cysteamine and exhibit a modified stress response in part due to enhanced intracellular gluthatione levels [11,14]. Indeed, cysteamine inhibits c-glutamylcysteine synthetase, the ratelimiting enzyme of gluthatione synthesis [15,16].…”
