PanIN-associated pericyte, glial, and islet remodeling in mice revealed by 3D pancreatic duct lesion histology

Abstract: Pericytes and glial cells are accessory cells of neurovascular networks, which have been reported to participate in scar formation after tissue injury. However, it remains unclear whether similar reactive cellular responses occur in pancreatic intraepithelial neoplasia (PanIN). In this study we developed three-dimensional (3D) duct lesion histology to investigate PanIN and the associated pericyte, glial, and islet remodeling. Transparent mouse pancreata with a Kras G12D mutation were used to develop 3D duct le… Show more

“…The technical advance of tissue clearing of mouse and human gastrointestinal tissues [10,37,[41][42][43] has enabled us to develop 3D histology for visualisation of the pancreatic neuroinsular network. Using the transparent specimens, we applied both fluorescence and transmitted light microscopy (a dualoptical approach) to examine the pancreatic microstructure and neurovascular network.…”

“…Importantly, with the addition of multicolor immunolabeling, the 3D relationships of normal ducts, precursor lesions, invasive carcinoma, immune cells, islets of Langerhans, vessels, and nerves should be visualizable. 10,15 Clearing methodologies have been described for 100 years. 7 The more recent addition of immunolabeling using fluorescent antibodies has opened the door to 3D microscopy.…”

“…Propidium iodide staining (50 μg/ml, Invitrogen) was performed to reveal the nuclei. The labelled specimens were immersed in the optical clearing aqueous solution (RapiClear 1.52 solution [18,19], SunJin Lab, Hsinchu, Taiwan) 1 day before being imaged via confocal microscopy.…”
