PAMAM dendrimer roles in gene delivery methods and stem cell research

Abstract: Nanotechnology has provided new technological opportunities, which could help in challenges confronting stem cell research. Polyamidoamine (PAMAM) dendrimers, a new class of macromolecular polymers with high molecular uniformity, narrow molecular distribution specific size and shape and highly functionalised terminal surface have been extensively explored for biomedical application. PAMAM dendrimers are also nanospherical, hyperbranched and monodispersive molecules exhibiting exclusive properties which make th… Show more

“…34 Several modifications have been made on PAMAM to improve its physicochemical and biological properties as a gene carrier into MSCs, including using RGD peptide and hyaluronic acid. 35,36 In a recent report, PAMAM G5 was conjugated to lauric, myristic, and palmitic fatty acids to evaluate their effects on the gene delivery activity of PAMAM in MSCs. The highest efficiency was obtained by the vectors containing the smallest hydrophobic chains.…”

“…However, the use of viral vectors in human clinical trials raises significant safety issues, such as potential immunogenicity and reversion to pathogenicity 6 7 . This has encouraged the development of non-viral vectors with better biosafety profiles, including nanocomplexes based on dendrimers 8 , lipids, 9 or polysaccharides 10 .…”

“…To circumvent this problem, neutrally charged particles or negative charged naked pDNA or RNA itself have been explored. Proton-sponge polymers, including PAMAM and PEI exhibit pKa values between physiological and lysosomal pH, causing osmotic swelling and rupture of the endosome membrane, which releases of the polyplexes into the cytosol [40,64]. And, the application of dendrimer reported the higher attachment and prolonged retention of therapeutics in the ECM.…”
