p63 Is Necessary for the Activation of Human Papillomavirus Late Viral Functions upon Epithelial Differentiation

Abstract: The late phase of the human papillomavirus (HPV) life cycle is linked to epithelial differentiation, and we investigated the factors that regulate this process. One potential regulator is p63, a member of the p53 family of proteins, which modulates epithelial development, as well as proliferation capability, in stem cells. In this study, we examined the role of p63 in the HPV life cycle using a lentiviral knockdown system for p63. In epithelial cells, the ⌬N truncated isoforms of p63 predominate, while the ful… Show more

“…Published data have defined ⌬Np63 as an induced target of E7, and E7 expression was sufficient for stimulating ⌬Np63 expression through inhibition of miR-203 microRNA production (23). Moreover, ⌬Np63 expression promoted productive viral DNA amplification (24). It is possible that E7 stimulates ⌬Np63 expression to support FA phenotypes and viral replication.…”

“…Lentivirus was produced as previously described (40). Plasmids carrying the short hairpin RNA (shRNA) sequences for Rad51 (TRCN0000018876 and TRCN0000018879) or BRCA1 (TRCN0000009824 and TRCN0000039833) or a scramble nontarget control shRNA cloned into the pLKO.1-puro background were obtained from the UNC Lentiviral Core Facility (Chapel Hill, NC).…”

“…Late gene expression is a second late viral event that is associated with differentiation status (4,28). Activation of the HPV-31 late promoter induces expression of transcripts encoding E1ˆE4, E1, E2, and E5 and can occur independently of amplification, which acts primarily to increase total transcript levels (28).…”
