P21/WAF1 is an independent survival prognostic factor for patients with hepatocellular carcinoma after resection

Abstract: In HCC, increased proliferation index PCNA is significantly associated with positive p53 and p21/WAF1. But p21/WAF1 expression did not relate to p53 expression. P21/WAF1 (+) is a good event and serves as an independent survival prognostic factor for HCC, which is a novel finding apart from previous reports.

“…Both hSSB1 and p21 were mainly located in the nuclei of the cancer cells and were highly expressed in the HCC tissues (Figure 6a). This result was consistent with the majority of previous studies showing that the tumor cells of HCC exhibited positive nuclear staining of p21 (Qin et al, 1998;SchonigerHekele et al, 2005;Kao et al, 2007;Lee et al, 2009). However, some reports have showed cytoplasmic p21 staining in HCC (Tanaka et al, 2002;Lee and Helfman, 2004;Shiraki and Wagayama, 2006), and we also observed a weak cytoplasmic p21 staining in three cases (3%) of HCC tissues that were excluded in our statistical analyses in this report.…”

“…p21 was found to be down-regulated in HCC tissues, demonstrating its tumor suppressive function [25][26][27] . Kao et al [28] also reported that p21 expression was observed in 37% of HCC tissues, regardless of p53 expression, and was an independent survival good prognosis factor. While most of the reports show that p21 acts as a tumor suppressor, expression levels of p21 in liver cirrhosis have been reported to be correlated with the cumulative incidence of the occurrence HCC [29] and to be dominant in cytoplasm when histology became more undifferentiated [30] .…”

“…The cyclin kinase inhibitor p21/ WAF1 is a key molecule in the regulation of cellular proliferation. A recent study by Kao et al [55] indicated that the immunopositivity for P21/WAF1 in tumorous tissue correlated with patient survival and served as an independent prognostic factor of survival after surgical resection for HCC. In addition, many other prognostic markers have been retrospectively evaluated for HCC patients, but randomized prospective studies are needed to corroborate robust prognostic markers.…”
