p21-activated kinase signalling in pancreatic cancer: New insights into tumour biology and immune modulation

Abstract: Pancreatic cancer is one of the most aggressive and lethal malignancies worldwide, with a very poor prognosis and a five-year survival rate less than 8%. This dismal outcome is largely due to delayed diagnosis, early distant dissemination and resistance to conventional chemo-therapies. Kras mutation is a well-defined hallmark of pancreatic cancer, with over 95% of cases harbouring Kras mutations that give rise to constitutively active forms of Kras. As important down-stream effectors of Kras, p21-activated kin… Show more

“…Pancreatic cancer is one of the malignant tumors with the worst prognosis. Due to its difficult early diagnosis and treatment, the 5‐year survival rate is less than 1% (Li et al, ; Wang, Baldwin, Nikfarjam, & He, ). Because pancreatic cancer is lack of obvious early symptoms, it is generally diagnosed in the advanced stages (Draper, ; Kitagawa et al, ).…”

“…The ve-year survival rate is less than 8% 1 . Surgery is the most important and comprehensive treatment for pancreatic cancer, and most patients have recurrence and metastasis earlier after operation.…”

“…The dectin1-galectin9 axis is central in directing the differentiation of TAM to a M2like phenotype, which suffices for reprogramming CD4+ and CD8+ T-cells (749). Finally, we list some reviews helpful as starting information on PaCa-selective metabolic changes that affect immune responses in PaCa (739,(750)(751)(752)(753)(754).…”
