P0 proteins of European beet-infecting poleroviruses display variable RNA silencing suppression activity

Abstract: Post-transcriptional gene silencing (PTGS), or RNA silencing, is one of the key mechanisms of antiviral defence used by plants. To counter this defence response, viruses produce suppressor proteins that are able to inhibit the PTGS pathway or to interfere with some of its function. The aim of this study was to evaluate the RNA silencing suppressor (RSS) activity of P0 proteins from selected European isolates of the beet-infecting poleroviruses beet chlorosis virus (BChV) and beet mild yellowing virus (BMYV) us… Show more

“…This is ORF0 encodes a putative protein of 28.1 kDa (P0). In other poleroviruses, this protein has RNA silencing suppressor activity [22][23][24] and we have also demonstrated this for TVDV P0 (Mo et al, unpublished results). It has been suggested that the P0 s of cucurbit aphid-borne yellow virus (CABYV) and turnip-yellow virus (TuYV) act as F-box proteins that target an essential component of the host post-transcriptional gene-silencing machinery [25].…”

“…This is also evidenced by P0 of different poleroviruses (Kozlowska-Makulska et al, 2010), as well as the P25 of several potexviruses (Senshu et al, 2009) that show different levels of RSS activity. The function of ADV P6 remains unknown.…”

“…Such isolate-dependent variability in the SSP activity was already observed for other poleroviral P0 proteins. For instance P0 encoded by some isolates of beet mild yellowing virus (BMYV) were reported to lack silencing suppression activity while other isolates of BMYV were fully competent in RNA silencing inhibition (Klein et al, 2014;Kozlowska-Makulska et al, 2010). Similarly Pfeffer et al (2002) reported a weak SSP activity for P0 encoded by a Dutch isolate of potato leafroll virus (PLRV), while Fusaro et al (2012) and Zhuo et al (2014) showed that P0 of an Australian and an Inner-Mongolian isolate respectively were strong inhibitors of RNA silencing.…”
