P‐0 Glycoprotein Overexpression Causes Congenital Hypomyelination Of Peripheral Nerves

Abstract: Abstract. We show that normal peripheral nerve myelination depends on strict dosage of the most abundantly expressed myelin gene, myelin protein zero ( Mpz ). Transgenic mice containing extra copies of Mpz manifested a dose-dependent, dysmyelinating neuropathy, ranging from transient perinatal hypomyelination to arrested myelination and impaired sorting of axons by Schwann cells. Myelination was restored by breeding the transgene into the Mpz -null background, demonstrating that dysmyelination does not result … Show more

“…Nevertheless, MPZ protein stimulation decreased after 72 h of treatment with LiCl (sevenfold stimulation). Such a discrepancy between MPZ transcript overexpression and protein decrease has already been described (12,13). We also observed that the increase in PMP22 The mean angular velocity of the whisker movements of all animals was analyzed by means of WhiskerWatcher software at DPI3, DPI8, and DPI14.…”
