Oxytocin and tumor necrosis factor α stimulate expression of prostaglandin E2 synthase and secretion of prostaglandin E2 by luminal epithelial cells of the porcine endometrium during early pregnancy

Abstract: Oxytocin (OXT) and tumor necrosis factor a (TNF) have been implicated in the control of luteolysis by stimulating endometrial secretion of luteolytic prostaglandin F 2a (PGF 2a ). Nevertheless, OXT concentration in porcine uterine lumen increases markedly on days 11-12 of pregnancy, and TNF is expressed in endometrium during pregnancy. The objective of the study was to determine the effect of OXT and TNF on expression of the enzymes involved in PG synthesis: PG-endoperoxide synthase 2 (PTGS2), PGE 2 synthase … Show more

“…PGE 2 , after reaching the ovaries, may act through luteal PGE 2 receptors (PTGER), PTGER2 and PTGER4 (Waclawik et al 2010). Other subtypes of PTGER have not been studied in the CL in this species yet.…”

“…The source of TNF may be macrophages, which are present in the endometrium during the estrous cycle and early pregnancy, as well as endometrial cells and conceptus (Hunt et al 1996). Recent studies indicate that TNF stimulates PGE 2 synthesis and secretion through upregulation of PTGS2 mRNA, as well as mPGES-1 mRNA and protein content in porcine LE cells collected from gilts on days 11-12 after estrus (Waclawik et al 2010).…”

“…Oestrogens and ovarian progesterone make up the specific hormonal microenvironment (Bazer and Johnson, 2014). The first oestrogens peak (days 10 to 12 of gestation) stimulates prostaglandin E2 (PGE2) synthesis and increases the PGE2/ prostaglandin F2α (PGF2α) ratio (Waclawik et al, 2009), boosts endometrial production of fibroblast growth factor 7 (FGF7), transforming growth factor beta (TGFβ), interleukin-1β, tumour necrosis factor alpha (TNFα), interferon delta (IFND) and interferon gamma (IFNG) (Harney and Bazer, 1989;Ka et al, 2001;Ross et al, 2003;White et al, 2009;Waclawik et al, 2010). The discussed growth factors and cytokines directly influence various genes whose products deliver luteoprotective effects and support conceptus development (Bazer and Johnson, 2014).…”
