Cuprous oxide (Cu2O) has great potential in photodynamic therapy for implant-associated infections due to its good biocompatibility and photoelectric properties. Nevertheless, the rapid recombination of electrons and holes weakens its photodynamic antibacterial effect. In this work, a new nanosystem (Cu2O@rGO) with excellent photodynamic performance was designed via the in situ growth of Cu2O on reduced graphene oxide (rGO). Specifically, rGO with lower Fermi levels served as an electron trap to capture photoexcited electrons from Cu2O, thereby promoting electron-hole separation. More importantly, the surface of rGO could quickly transfer electrons from Cu2O owing to its excellent conductivity, thus efficiently suppressing the recombination of electron-hole pairs. Subsequently, the Cu2O@rGO nanoparticle was introduced into poly-L-lactic acid (PLLA) powder to prepare PLLA/Cu2O@rGO porous scaffolds through selective laser sintering. Photochemical analysis showed that the photocurrent of Cu2O@rGO increased by about two times after the incorporation of GO nanosheets, thus enhancing the efficiency of photogenerated charge carriers and promoting electron-hole separation. Moreover, the ROS production of the PLLA/Cu2O@rGO scaffold was significantly increased by about two times as compared with that of the PLLA/Cu2O scaffold. The antibacterial results showed that PLLA/Cu2O@rGO possessed antibacterial rates of 83.7% and 81.3% against Escherichia coli and Staphylococcus aureus, respectively. In summary, this work provides an effective strategy for combating implant-related infections.

Photodynamic therapy (PDT) presented tremendous potential for combating implant-related infections, but its antibacterial efficacy was constrained by rapid electron−hole pair recombination of photosensitizers and barrier action of a dense biofilm on reactive oxygen species (ROS). Herein, a cascade phototherapeutic AuKN-LA nanosystem was constructed by depositing Au nanoparticles and L-arginine (LA) on potassium niobate (KN) photosensitizer. In the nanosystem, Au nanoparticles with lower Fermi levels could capture photogenerated electrons and thus suppress electron−hole pair recombination to enhance ROS generation. Especifically, L-arginine could cascade trigger by the generated ROS to release nitric oxide (NO) to destroy the biofilm, thereby facilitating the entry of ROS. Furthermore, NO itself possessed antibacterial activity, which could form synergy with PDT. Then, the nanosystem was introduced into poly(L-lactic acid) scaffolds fabricated by laser additive manufacturing. Photoelectrochemical and ROS analysis proved that the electron−hole pair separation was improved, significantly increasing ROS generation. NO detection and crystal violet staining showed that the scaffolds could effectively remove the biofilm by sustainably releasing NO under light irradiation. As a consequence, the scaffolds exhibited excellent antibacterial rates of 98.7% and 99.2% against E. coli and S. aureus, respectively, by disrupting bacterial cell membranes and causing leakage of nucleic acid molecules and proteins.

Poly methyl methacrylate (PMMA) bone cement is widely used in orthopedic surgeries, including total hip/knee arthroplasty and vertebral compression fracture treatment. However, loosening due to bone resorption is a common mid-tolate complication. Therefore, developing bioactive bone cement that promotes bone growth and integration is key to reducing aseptic loosening. In this study, we developed a piezoelectric bone cement comprising PMMA and BaTiO 3 with excellent electrobioactivity and further analyzed its ability to promote bone integration. Experiments demonstrate that the PMMA and 15 wt % BaTiO 3 cement generated an open-circuit voltage of 37.109 V under biomimetic mechanical stress, which effectively promoted bone regeneration and interfacial bone integration. In vitro experiments showed that the protein expression levels of ALP and RUNX-2 in the 0.65 Hz and 20 min group increased by 1.74 times and 2.31 times. In vivo experiments confirmed the osteogenic ability of PMMA and 15 wt % BaTiO 3 , with the increment of bone growth in the non-movement and movement groups being 4.67 and 4.64 times, respectively, at the second month after surgery. Additionally, Fluo-4 AM fluorescence staining and protein blotting experiments verified that PMMA and 15 wt % BaTiO 3 electrical stimulation promoted osteogenic differentiation of BMSCs by activating calcium-sensitive receptors and increasing calcium ion inflow by 1.41 times when the stimulation reached 30 min. Therefore, piezoelectric bioactive PMMA and 15 wt % BaTiO 3 cement has excellent application value in orthopedic surgery systems where stress transmission is prevalent.
