Oxidative Stress Is Involved in the Pathogenesis of Keshan Disease (an Endemic Dilated Cardiomyopathy) in China

Abstract: Oxidative stress and selenoprotein deficiency are thought to be associated with the pathogenesis of Keshan disease (KD). However, to our knowledge, the level of oxidative stress and expression of selenoproteins have not been investigated in the myocardium of patients with KD. In this study, 8-hydroxy-2-deoxy guanosine (8-OH-dG), a marker of oxidative stress, was used to assess the level of oxidative stress, and thioredoxin reductase 1 (TrxR1) and glutathione peroxidase 1 (GPx1) were assessed to reflect the lev… Show more

“…The average integrated optical density (IOD) of SOCS3 IHC staining in CD and normal control tissues was calculated with Image-Pro Plus software (version 5.0) as previously reported 34 . Five fields were analysed on each slide 35 .…”

“…Sections incubated with PBS instead of the primary antibody were chosen as negative controls. Ten fields of each section were randomly selected and microscopically examined, and stathmin immunoreactivity was analyzed by measuring the integrated optical density (IOD) by Image pro plus 6.0 (Media Cybernetics, Silver Spring, MD), as described previously . After the calibration of optical density, we used Hue Saturation Intensity mode (hue = 0–30; saturation = 0–255; intensity = 0–230) to select the area of interest, and the IOD that indicated stathmin immunoreactivity were counted.…”

“…Several studies on heart failure have provided consequential reports that oxidative stress enhances under the conditions of heart failure and contributes to disease progression [73]. The pivotal role of oxidative stress is also well explained in diabetic cardiomyopathy, endemic cardiomyopathy (Keshan disease) and Chagas disease [74][75][76][77]. The cardiomyopathy includes, among others, asymptomatic diastolic dysfunction and end-stage left ventricular (LV) dilation with reduced systolic function.…”
