Overview of methodologies for T-cell receptor repertoire analysis

Abstract: BackgroundThe T-cell receptor (TCR), located on the surface of T cells, is responsible for the recognition of the antigen-major histocompatibility complex, leading to the initiation of an inflammatory response. Analysing the TCR repertoire may help to gain a better understanding of the immune system features and of the aetiology and progression of diseases, in particular those with unknown antigenic triggers. The extreme diversity of the TCR repertoire represents a major analytical challenge; this has led to t… Show more

“…A further step forward came with the use of template switch enzymes and 5′ RACE, as has been frequently used in T cell biology 59, 60, 61, 62, 63, 64, 65. The 5′ RACE method has an advantage over consensus immunoglobulin PCR because it only requires priming in the constant region and adds a primer landing site in the 5′ with the addition of a template switch oligo (TSO).…”

“…The assessment of TCR repertoire diversity is becoming increasingly accessible [219] . Important choices such as library preparation method, in-house versus service provider, output data type (raw and/or analyzed), and the use or not of unique molecular identifiers must first be matched to project goals [220] . Basic features of the T cell receptor repertoire are still being revealed, e.g.…”
