Overexpression of suppressive microRNAs, miR-30a and miR-200c are associated with improved survival of breast cancer patients

Abstract: Some microRNAs (miRNAs) are known to suppress breast cancer. However, whether the expressions of these tumor suppressive miRNAs translate to patient survival were not investigated in large cohort. Nine miRNAs (miR-30a, miR-30c, miR-31, miR-126, miR-140, miR-146b, miR-200c, miR-206, and miR-335) known to be tumor suppressive miRNAs in breast cancer were investigated in Genomic Data Common data portal miRNA-Seq dataset and The Cancer Genome Atlas (TCGA) (n = 1052). Of the 9 miRNAs, miR-30a, miR-30c, miR-126, miR… Show more

“…netDx selected pathways with themes similar to the Luminal A gene expression‐based predictor previously mentioned (Fig B), including cell cycle regulation and DNA damage repair (Appendix Fig S4A). DIABLO identified a range of features correlated with outcome, such as the known link of mir‐30a overexpression with improved survival in both luminal and basal tumors (Kawaguchi et al , ). It also highlighted individual genes present in the pathways found by netDx (Appendix Fig S4B), for example, CENPA and AURKB in Aurora signaling pathways.…”

“…In previous studies, a substantial number of drug‐metabolizing enzymes under the regulatory control of the AhR signaling pathway have been shown to be modulated by miRNAs and certain miRNAs have been shown to be differently expressed in cancer tissues in comparison to normal tissues . Also the distinct role of estrogen receptor in developing cancer has been documented so far .…”

“…Another extensively investigated miRNA is miR‐206, which regulates cell cycle arrest and inhibits estrogen‐induced cell proliferation . miR‐206 also suppresses cell migration in breast cancer cells by the inhibition of the ERα expression . In accordance, the expression of miR‐133a is downregulated in breast cancer cells.…”

“…[25,26]. Its upregulation, nevertheless, was associated with increased sensitivity to radiation treatment, and it is associated with better overall survival in ER positive breast cancer [27,28]. miR-200c belongs to a cluster of miRNAs known as the miR-200bc/429 cluster.…”
