Overexpression of SmANS Enhances Anthocyanin Accumulation and Alters Phenolic Acids Content in Salvia miltiorrhiza and Salvia miltiorrhiza Bge f. alba Plantlets

Abstract: Flavonoids play multiple roles in plant coloration and stress resistance and are closely associated with human health. Flavonoids and non-flavonoids (such as phenolic acids) are produced via the phenylpropanoid-derived pathway. Anthocyanidin synthase (ANS) catalyzes the synthesis of anthocyanins from leucoanthocyanidin in the flavonoids branched pathway. In this study, SmANS from Salvia miltiorrhiza was cloned and mainly localized in the endoplasmic reticulum (ER), plastids, Golgi, plasma membrane, and nucleus… Show more

“…The suppressed expression of the ANS gene leads to a lack of anthocyanins in Caryophyllales [ 35 ]. Li et al [ 36 ] reported that overexpression of SmANS increased anthocyanin content but reduced the biosynthesis of salvianolic acid B in S. miltiorrhiza. White-flowered S. miltiorrhiza is a variety of the S. miltiorrhiza species.…”

“…Moreover, salvianolic acid B has a high concentration in the liver and a transporter known as OATP1B1 is involved in its hepatic uptake and clearance [22]. In general, flavonoids are implicated in plant coloration and stress resistance [23].…”

“…The formula A 530 ‐0.25(A 657 ) was used to measure the absorbance values (A) of the extract at 530 and 657 nm. Three biological replicates per treatment were used to analyze anthocyanin levels as previously described (Li et al 2019).…”

“…A series of flavonoids are generated from 4‐coumaroyl‐CoA via pathways catalyzed by key enzymes such as chalcone synthase (CHS), chalcone isomerase (CHI), flavanone 3‐hydroxylase (F3H), dihydroflavonol reductase (DFR), anthocyanidin synthase (ANS), and others (Figure S2; Li et al 2019; Ni et al 2019). Identifying these biosynthetic pathway genes has laid the foundation for in‐depth genetic engineering studies.…”
